circ_0007841靶向miR-513a-5p对胃癌细胞增殖、侵袭和迁移的影响

摘要 目的:探讨circ_0007841靶向miR-513a-5p对胃癌细胞增殖、侵袭和迁移的影响。方法:RT-qPCR检测胃癌组织、癌旁组织、胃癌细胞HGC-27以及人胃黏膜细胞GES-1中circ_0007841和miR-513a-5p表达。双荧光素酶报告实验确定circ_0007841和miR-513a-5p相互作用。将HGC-27细胞分为si-NC组、si-circ_0007841组、miR-NC组、miR-513a-5p mimic组、si-circ_0007841+miR-513a-5p Inhibitor组。CCK-8法检测HGC-27细胞存活率;平板克隆实验检测HGC-27细胞克隆形成数;划痕愈合实验和Transwell实验检测HGC-27细胞迁移能力;Transwell实验检测HGC-27细胞侵袭能力。结果:与癌旁组织比较,胃癌组织中circ_0007841相对水平显著升高(P<0.05),miR-513a-5p相对水平显著降低(P<0.05)。与GES-1细胞比较,HGC-27细胞中circ_0007841相对水平显著升高(P<0.05),miR-513a-5p相对水平显著降低(P<0.05)。circ_0007841和miR-513a-5p直接特异性结合。与si-NC组比较,si-circ_0007841组HGC-27细胞存活率、克隆形成数、划痕愈合率、迁移数、侵袭数显著降低(P<0.05),miR-513a-5p相对水平显著升高(P<0.05)。与miR-NC组比较,miR-513a-5p mimic组HGC-27细胞存活率、克隆形成数、划痕愈合率、迁移数、侵袭数显著降低(P<0.05)。与si-circ_0007841组比较,si-circ_0007841+miR-513a-5p Inhibitor组HGC-27细胞存活率、克隆形成数、划痕愈合率、迁移数、侵袭数显著升高(P<0.05)。结论:干扰circ_0007841通过靶向上调miR-513a-5p表达来抑制胃癌细胞增殖、侵袭和迁移。 Objective:To explore the role of circ_0007841 on cell proliferation,invasion and migration in gastric cancer through targeting miR-513a-5p.Methods:Expression of circ_0007841 and miR-513a-5p in gastric cancer tissues,adjacent tissues,gastric cancer cells HGC-27 and human gastric mucosal cells GES-1 was detected by RT-qPCR.Dual luciferase reporter assay was used to confirm the interaction between circ_0007841 and miR-513a-5p.HGC-27 cells were divided into si-NC group,si-circ_0007841 group,miR-NC group,miR-513a-5p mimic group,si-circ_0007841+miR-513a-5p Inhibitor group.CCK-8 method detected the survival rate of HGC-27 cells;the clone formation number of HGC-27 cells was determined by plate cloning assay;the migration ability of HGC-27 cells was measured by scratch healing and the Transwell assays;the invasion ability of HGC-27 cells was tested using Transwell assay.Results:Compared with adjacent tissues,the relative level of circ_0007841 in gastric cancer tissue was markedly increased(P<0.05),while the relative level of miR-513a-5p was markedly decreased(P<0.05).Compared with GES-1 cells,the relative level of circ_0007841 in HGC-27 cells was markedly increased(P<0.05),while the relative level of miR-513a-5p was markedly decreased(P<0.05).circ_0007841 directly and specifically bound to miR-513a-5p.Compared with the si-NC group,the survival rate,clone formation numbers,wound healing rate,migration numbers and invasion numbers of HGC-27 cells in the si-circ_0007841 group were markedly reduced(P<0.05),and the relative level of miR-513a-5p was markedly increased(P<0.05).Compared with the miR-NC group,the survival rate,clone formation numbers,wound healing rate,migration numbers and invasion numbers HGC-27 cells in the miR-513a-5p mimic group were markedly reduced(P<0.05).Compared with the si-circ_0007841 group,the survival rate,clone formation numbers,wound healing rate,migration numbers and invasion numbers HGC-27 cells in the si-circ_0007841+miR-513a-5p Inhibitor group were markedly increased(P<0.05).Conclusion:Interference circ_0007841 inhibits cell proliferation,invasion and migration in gastric cancer by targeting and up-regulating miR-513a-5p expression.
