姜黄素调控TLR4/mTOR信号通路对肝癌细胞增殖、侵袭和迁移的作用

Effect of curcumin on the proliferation,invasion and migration of hepatocarcinoma cells by regulating TLR4/mTOR signaling pathway

目的:分析姜黄素影响肝癌(LC)细胞系BEL-7402体外侵袭、转移与增殖及其潜在分子机制。方法:对BEL-7402细胞实施浓度不等的姜黄素(0、30、60μmol/L)干预,经由四甲基偶氮唑蓝(MTT)技术对细胞的增殖情况展开测定,Transwell法对细胞侵袭与迁移活性展开测定;用姜黄素(60μmol/L)处理BEL-7402细胞,Western blot法检测细胞内Toll样受体4(TLR4)/哺乳动物雷帕霉素靶蛋白(mTOR)表达情况。结果:MTT实验显示,在BEL-7402细胞增殖能力上,相较空白对照组,姜黄素1组、2组皆显著偏低(P<0.05)。Transwell侵袭和迁移实验结果显示,相较于空白对照组,姜黄素1组和姜黄素2组BEL-7402细胞侵袭能力和迁移能力均显著降低(P<0.05)。Western blot结果表明,姜黄素2组BEL-7402内mTOR、TLR4蛋白表达量显著下调(P<0.05)。结论:对于LC细胞BEL-7402的增殖、转移与侵袭,姜黄素具抑制功能,其机制和下调信号途径TLR4/mTOR可能相关。

Objective:To investigate the effect of curcumin on the proliferation,migration and invasion of hepatocellular carcinoma cell line BEL-7402 in vitro and its potential molecular signaling mechanism.Methods:BEL-7402 cells were treated with different concentrations of curcumin(0,30,60μmol/L).The proliferation of BEL-7402 cells was detected by MTT assay,and the migration and invasion ability of BEL-7402 cells were detected by Transwell assay.BEL-7402 cells were treated with curcumin(60μmol/L),and the expression of Toll-like receptor 4(TLR4)/mammalian target of rapamycin(mTOR)was detected by Western blot.Results:MTT test showed that BEL-7402 cell proliferation ability in curcumin 1 and curcumin 2 groups was significantly decreased compared with blank control group(P<0.05).The results of Transwell invasion and migration experiment showed that the invasion and migration ability of BEL-7402 cells in curcumin 1 and curcumin 2 groups were significantly decreased compared with blank control group(P<0.05).Western blot results showed that in curcumin 2 group,the expression levels of TLR4 and mTOR proteins in BEL-7402 cells were significantly decreased(P<0.05).Conclusion:Curcumin can inhibit the proliferation,invasion and migration of BEL-7402 cells,and the mechanism may be related to down-regulation of TLR4/mTOR signaling pathway.
