地卷属地衣1中国新记录种——栗色地卷(新拟)

摘要 该研究以采集于河北省小五台山自然保护区地卷属地衣为试验材料,通过形态解剖学、地衣次生代谢产物和nrDNA的ITS分子系统学分析,对地卷属地衣进行鉴定。结果表明:(1)发现小五台山分布的地卷属1中国地衣新记录种——栗色地卷(新拟)(Peltigera castanea Goward,Goffinet&Miadl.)。(2)该种的识别特征为:地衣体上表面灰褐色至暗褐色,中部稍具光泽;裂片顶端边缘稍具绒毛;粉芽颗粒状,褐色,并聚集成不规则圆形;下表面脉纹扁平,淡白色、淡褐色至淡黑色;假根密集,束状、丛生,上部呈柔毛状。(3)该种标本ITS序列与已经提交的栗色地卷序列的一致性为98.3%~99.1%,且系统发育树分析显示,该种ITS序列与GenBank中Peltigera castanea的ITS序列聚为一支,并具有95%/99%的支持强度,故确定为地卷属地衣栗色地卷(新拟)。标本存放于中国科学院微生物研究所真菌标本馆地衣标本室(HMAS-L)。 In this study,lichens of the genus Peltigera Willd.collected from Xiaowutai Mountain Nature Reserve in Hebei Province are used as experimental materials to identify the species of Peltigera Willd.by means of morphological anatomy,detection of lichen secondary metabolites and ITS sequence of nrDNA.The results show that:(1)Peltigera castanea Goward,Goffinet&Mi dl.is reported new to China,which is found in Mt.Xiaowutai of Hebei Province.(2)The newly recorded species can be distinguished by upper surface grey brown to dark brown,more or less shiny near the central part and lobes tips slightly tomentose.Soredium is granular,brown,and gathered into an irregular circle.The veins on the lower surface are flat,pale white,pale brown or pale black.The rhizoids are dense,fasciculate and tufted,becoming flocculent near rhizoid tips.(3)The consistency between the ITS sequence of this specimen and the previously submitted P.castanea sequence is 98.3%-99.1%,and phylogenetic tree analysis shows that this ITS sequence is clustered with the ITS sequence of P.castanea in GenBank,with a support strength of 95%/99%.Therefore,it is determined as P.castanea.The specimens are deposited in the Herbarium Mycologicum Academiae Sinicae-Lichenes(HMAS-L).
