摘要
精原干细胞(SSCs)作为成体干细胞之一,可将遗传信息向子代传递,在雄性动物的精子发生过程中起到至关重要的作用。根据SSCs的生物学特征并通过有效的细胞分离手段将睾丸中的SSCs进行分离,进一步纯化并体外培养,才能更加深入地研究其相关机制。随着测序技术及基因编辑技术的发展和成熟,对于SSCs的研究手段和应用范围也在不断扩展。因此,文章主要对SSCs的生物学特征、鉴定、分离、纯化、体外培养以及与SSCs研究中联合使用的新技术进行了总结,以期为SSCs体外培养方法选择及其相关研究提供参考。
As one of the adult stem cells,SSCs can transmit genetic information to offspring and play a crucial role in the spermatogenesis of male animals.According to the biological characteristics of SSCs and effective cell separation methods,SSCs in testis can be separated and further purified and cultured in vitro,so as to further study their related mechanisms.With the development and maturity of sequencing technology and gene editing technology,the research means and application range of ssCs are also expanding.Therefore,the paper mainly summarizes the biological characteristics,identification,isolation,purification,and in vitro culture of SSCs,as well as the new technologies used in combination with SSCs research,in order to provide references for the selection of in vitro culture methods of SSCs and related studies.
