青花椒精油柔性脂质体的制备及质量评价

Preparation of Flexible Nanoliposomes of Volatile Oil from Zanthoxylum schinifolium and Its Quality Evaluation

摘要 制备青花椒精油柔性脂质体,并考察其在小鼠体外的透皮吸收特征。采用薄膜-超声法,以包封率和粒径为指标,泊洛沙姆188为表面活性剂制备青花椒精油柔性脂质体,并对制备成功的柔性脂质体进行质量评价。通过优选制得的青花椒精油柔性脂质体平均粒径为115.81±2.11 nm(n=3),包封率为92.13%±2.77%,PDI为0.236±0.110(n=3),Zeta电位为-50.21±2.35 mV(n=3),纳米粒积累渗透量为149.24μg/cm^(2),是原料的1.77倍。该青花椒精油柔性脂质体制备工艺方便科学、稳定可行,粒径小且分布均匀,能够增加青花椒精油的经皮渗透,是一种良好的经皮给药载体。 Prepare flexible liposomes of volatile oil from Zanthoxylum schinifolium,and investigate its properties and transdermal absorption behavior characteristics of its mouse skin in vitro.Poloxam 188 was prepared as flexible liposome for the volatile oil.On the basis,the flexible liposomes of volatile oil from Zanthoxylum schinifolium were prepared by membraneultrasonic method with entrapment efficiency and size as index,and the quality was evaluated.The mean particle size,entrapment efficiency,PDI and Zeta potential of the flexible liposomes of volatile oil from Zanthoxylum schinifolium prepared by optimization were 115.81±2.11 nm(n=3),92.13%±2.77%,0.236±0.110(n=3)and-50.21±2.35 mV(n=3),respectively.The accumulation and permeability of liposomes reached 149.24μg/cm^(2),1.77 times of the raw material.The flexible liposomes had small particle size,even distribution and high entrapment efficiency.Its preparation process of flexible liposomes is convenient,scientific,stable and feasible,and can promote the transdermal penetration of volatile oil from Zanthoxylum schinifolium,thus being a good transdermal drug delivery carrier.
