摘要
目的 研究四方木皮化学成分及抗炎镇痛的作用。方法 利用UPLC-MS/MS技术定性分析四方木皮次生代谢成分,并借助TCMSP数据库平台分析其活性成分对应靶点和疾病。采用小鼠耳肿胀法、足趾肿胀法研究四方木皮的抗炎作用;热板法、扭体实验研究四方木皮的镇痛作用。结果 四方木皮共鉴定出7类541种次生代谢物,其中,TCMSP平台鉴定出120种中药化学成分,52种成分与抗炎镇痛显著相关,其中数量较多的是黄酮类与酚酸类代谢物;四方木皮低、中、高剂量组均可明显抑制小鼠耳肿胀和足趾肿胀以及减少小鼠的扭体次数(P <0.05);与生理盐水组相比,四方木皮能有效提高小鼠的痛阈值,但差异无统计学意义(P> 0.05)。结论 四方木皮乙醇提取物具有一定抗炎镇痛作用,黄酮和酚类物质在四方木皮的中药活性及抗炎镇痛方面中起重要作用,为进一步阐明四方木皮的抗炎、镇痛作用机制提供了基础。
Objective To study the chemical constituents and anti-inflammatory and analgesic effects of Saracae Cortex.Methods UPLC-MS/MS technology was used to qualitatively analyze the secondary metabolites of Saracae Cortex,and TCMSP database platform was used to analyze the corresponding targets and diseases of its active components.The anti-inflammatory effect of Saracae Cortex was observed by ear swelling and toe swelling in mice.The analgesic effect of quadrilateral bark was observed by hot plate method and writhing test.Results A total of 541 secondary metabolites in 7 categories were identified from Saracae Cortex.Among them,120 chemical components of traditional Chinese medicine were identified by TCMSP platform,and 52 substances were significantly related to anti-inflammatory and analgesic effects.The low,medium and high dose groups could significantly inhibit ear swelling and toe swelling and reduced the number of writhing in mice(P<0.05).Compared with the saline group,Saracae Cortex could effectively improve the pain threshold of mice,but the difference was not statistically significant(P>0.05).Conclusion Saracae Cortex has certain anti-inflammatory and analgesic effects.Flavonoids and phenols play an important role in the traditional Chinese medicine activity and anti-inflammatory and analgesic effects of Saracae Cortex,which provides a certain research direction and data support for further elucidating the anti-inflammatory and analgesic mechanisms of Saracae Cortex.
