皖南地区汉族人群C3435T位点基因多态性与卒中后抑郁的关联分析

Analysis of gene polymorphism at the C3435T locus with post-stroke depression in a Han Chinese population in southern Anhui Province

目的探究皖南地区汉族人群中ABCB1 C3435T位点基因多态性与卒中后抑郁(post-stroke depression,PSD)的关联,并分析其相关性。方法采用回顾性研究方法,研究对象为2018年1月至2022年6月于我院就诊的228例患者,使用汉密尔顿抑郁量表(Hamilton Depression Scale,HAMD)进行评估,以确定抑郁症严重程度,并完成分组。其中卒中后抑郁组(PSD组)96例,非抑郁组(NPSD组)132例。采用荧光染色原位杂交及染色体核型分析系统完成入组患者ABCB1 C3435T位点基因分型。通过强迫引入法分析C3435T位点基因多态性与卒中后抑郁的关联,并分析分布频率和相对危险度。结果PSD组次要等位基因频率为47.9%,高于NPSD组,以等位基因C为参照,T的相对危险度为1.67;以CC基因型为参照,CT、TT的OR值分别为1.938、2.527,差异有统计学意义。结论皖南地区汉族人群中ABCB1 C3435T位点基因多态性与PSD之间具有相关性,等位基因T增加PSD的发病风险,未来仍需进一步研究去深入探讨。

Objective To investigate the association between gene polymorphisms at the ATP-binding cassette subfamily B member 1(ABCB1)C3435T locus and post-stroke depression(PSD)in the Han population in southern Anhui Province.Methods Using a retrospective study method,228 patients who attended our hospital from January 2018 to June 2022 were assessed using the Hamilton Depression Scale(HAMD)to determine the grouping by the degree of depression.There were 96 cases in the PSD group and 132 cases in the non-depressed group(NPSD).Genotyping of the C3435T locus was completed using fluorescent staining in situ hybridization and a karyotype analysis system.The association between genetic polymorphisms at the C3435T locus and post-stroke depression was analyzed by the forced introduction method,and the distribution frequency and relative risk were analyzed.Results The frequency of minor alleles was 47.9%in the PSD group,which was higher than that in the NPSD group,and the relative risk of T was 1.67 with allele C as a reference.The ORs of CT and TT were 1.938 and 2.527 with CC genotypes as reference,respectively,which were statistically significant.Conclusion There is a correlation between gene polymorphism at the ABCB1 C3435T locus and PSD in the Han population in southern Anhui.And allele T increases the risk of developing PSD still needs further studies to explore in the future.
