胆管癌免疫细胞浸润模式及预后相关性分析

Analysis of immune cell infiltration pattern and prognosis of cholangiocarcinoma

目的研究胆管癌组织的免疫细胞浸润的特征及其与临床预后之间的相关性。方法从癌症基因组图谱(the cancer genome atlas,TCGA)数据库中下载胆管癌组织和癌旁组织的转录本数据及临床信息数据,用反卷积算法(CIBERSORT)分析胆管癌组织及癌旁组织中22种肿瘤浸润性免疫细胞(tumorinfiltrating immune cells,TIICs)的比例。采用R软件包绘制免疫细胞相关性热图及条形图。利用R软件包对胆管癌免疫细胞浸润模式与胆管癌的生存时间进行分析,用条形图展示TIICs在胆管癌组织中的比例,使用小提琴图和热图展示免疫细胞在胆管癌和癌旁组织中表达的区别。结果从TCGA数据库中,共获取了17例胆管癌组织和3例癌旁组织,分析后发现,免疫浸润细胞在胆管癌组织中的比例不同。在提取的细胞亚群中,T细胞,NK细胞,单核细胞和巨噬细胞比例较高,而B细胞和中性粒细胞的所占比值相对较低。生存分析结果显示,胆管癌组织中不同的TIICs比例和患者的生存时间相关。高构成比的浆细胞提示较差的临床预后(P<0.05)。结论胆管癌组织TIICs亚群比例的不同和临床特征密切相关,可为临床预后评价提供参考依据。

Objective To investigate the characteristics of immune cell infiltration in cholangiocarcinoma tissues and its correlation with clinical prognosis.Methods Transcript data and clinical information data of cholangiocarcinoma tissues and paraneoplastic tissues were downloaded from the cancer genome atlas(TCGA) database,and 22 types of tumor infiltrating immune cells(TIICs) in cholangiocarcinoma tissues and paraneoplastic tissues were analyzed by the inverse convolution algorithm(CIBERSORT).Immune cell correlation heat map and bar graph were plotted using R package.The R package was used to analyze the pattern of immune cell infiltration in cholangiocarcinoma and survival time in cholangiocarcinoma,to demonstrate the proportion of TIICs in cholangiocarcinoma tissues using bar graphs,and to demonstrate the difference in immune cell expression in cholangiocarcinoma and paraneoplastic tissues using violin plots and heat maps.Results From the TCGA database,a total of 17 bile duct cancer tissues and 3 paraneoplastic tissues were obtained,and the analysis revealed that the proportion of immune infiltrating cells differed in bile duct cancer tissues.Among the extracted cell subpopulations,T cells,NK cells,monocytes and macrophages had a high proportion,while the ratio of B cells and neutrophils was relatively low.Survival analysis showed that the different proportions of TIICs in cholangiocarcinoma tissues correlated with the survival time of patients.The high composition ratio of plasma cells suggested a poorer clinical prognosis(P<0.05).Conclusion The different proportions of TIICs subpopulations in cholangiocarcinoma tissues are highly correlated with clinical characteristics,which can provide a reference for clinical prognosis evaluation.
