摘要
目的探究p53基因突变对胶质母细胞瘤细胞连接蛋白43(Connexin 43)表达的影响。方法培养胶质母细胞瘤U87细胞(p53野生型)与U251细胞(p53突变型),采用Western blotting方法分别检测两种细胞中p53蛋白与Connexin 43蛋白的表达水平。结果胶质母细胞瘤U251细胞中p53基因发生突变,与U87细胞相比,p53蛋白表达水平降低(t=2.948,P<0.05);U251细胞中Connexin 43蛋白的表达水平显著高于U87细胞(t=2.771,P<0.05)。结论在胶质母细胞瘤细胞中,p53基因突变可能会提高Connexin 43蛋白的表达量。
Objective To investigate the effects of p53 gene mutation on the expression of Connexin 43 in glioblastoma cells.Methods In cultured glioblastoma U87 cells(wild-type p53 gene)and U251 cells(mutant p53 gene),the expression of p53 protein and Connexin 43 protein was measured by Western blot.Results The p53 protein level was significantly decreased in U251 cells compared with U87 cells(t=2.948,P<0.05).The expression level of Connexin 43 protein in U251 cells was significantly increased than that of U87 cells(t=2.771,P<0.05).Conclusion The mutation of the p53 gene may increase the expression of Connexin 43 protein in glioblastoma cells.
