苹果砧木‘SH6’叶绿体基因组序列特征和比较分析

Sequence and comparative analysis of chloroplast genome of apple rootstock‘SH6’

【目的】为了解析苹果矮化中间砧‘SH6’叶绿体基因组特征及其进化关系。【方法】以苹果矮化中间砧‘SH6’为试材,通过HIFI测序,利用Organelle_PBA软件进行叶绿体基因组从头组装,分析其叶绿体基因组特征。【结果】结果表明,‘SH6’叶绿体基因组大小为160069 bp,具有典型的四分结构,包括大单拷贝区域、小单拷贝区域和两个反向重复区域,长度分别为88184 bp、19181 bp、26352 bp和26352 bp。在‘SH6’叶绿体基因组中,分别注释了蛋白质编码基因、tRNA和rRNA,其数量分别为86、36和8。此外,整合已发表的‘国光’、河南海棠、2个野生苹果和桃(Prunus persica)的叶绿体基因组数据来构建系统发育树,发现苹果中间砧‘SH6’与欧洲森林苹果聚在一枝上。【结论】推测苹果矮化中间砧品种‘SH6’与欧洲森林苹果关系较为密切。

[Objective]In order to analyze the chloroplast genome characteristics and evolutionary relationship of apple dwarf intermediate rootstock‘SH6’.[Methods]The apple dwarf intermediate rootstock‘SH6’were sequenced by HIFI,and the chloroplast genome was de novo assembled with the Organelle_PBA software to analysis its genomic features.[Results]The results showed that the chloroplast genome of‘SH6’was 160069 bp and had a typical tetrad structure including a large single copy region,a small single copy region and two reverse repeat regions with lengths of 88184 bp,19181 bp,26352 bp and 26352 bp.In the chloroplast genome of‘SH6’,protein coding genes,tRNA and rRNA were annotated,and their numbers were 86,36 and 8,respectively.In addition,a phylogenetic tree was constructed with the published chloroplast genomes of‘Rall’,Malus honanensis,two wild apples,and Prunus persica,and it was found that the apple intermediate anvil‘SH6’clusters with apple of M.sylvestris.[Conclusion]The apple dwarf intermediate rootstock‘SH6’is relatively closely related to M.sylvestris.
