摘要
目的:建立反相高效液相色谱(reversed-phase high performance liquid chromatogra,RP-HPLC)法测定香青兰乙酸乙酯提取物中木犀草素-7-O-β-D-葡萄糖醛酸苷、迷迭香酸和田蓟苷的含量。方法:采用Inertsil ODS-SP(4.6×150 nm,5μm)色谱柱,以乙腈(A)-0.1%甲酸水溶液(B)为流动相,梯度洗脱,流速1 mL/min,检测波长254 nm,柱温35℃,进样体积10μL。结果:木犀草素-7-O-β-D-葡萄糖醛酸苷、迷迭香酸与田蓟苷分离效果良好,分别在10.6~106.0μg/mL(r=0.9987)、11.5~115.2μg/mL(r=0.9994)、8.1~81.2μg/mL(r=0.9996)呈良好的线性关系;平均回收率为97.83%、99.07%、98.63%;RSD值为1.03%、1.44%以及0.69%。结论:该方法重复性好,结果准确可靠,为香青兰提取物中木犀草素-7-O-β-D-葡萄糖醛酸苷、迷迭香酸和田蓟苷成分的含量测定提供科学依据。
Objective:To establish a RP-HPLC method for determination of Luteolin-7-O-β-D-Glucuronide,rosmarinic acid and tilianin in ethyl acetate extract of Mongolia Dracocephalum moldavica L.Methods:The test was conducted on chromatographic column of Inertsil ODS-SP(4.6×150nm,5 um),and the mobile phase was acetonitrile(A)-0.1%formic acid(B)for gradient elution.The flow rate was 1.0 mL/min,the detection wavelength was 254 nm and the column temperature was 35℃,the injection volumn 10μL.Results:Luteolin-7-O-β-D-Glucuronide,rosmarinic acid and tilianin displayed good linear relationships within the ranges of 10.6~106.0μg/mL(r=0.9987),11.5~115.2μg/mL(r=0.9994),and 8.1~81.2μg/mL(r=0.9996),and the average recoveries were 97.83%,99.07%,and 98.63%,with the RSDs of 1.03%,1.44%,and 0.69%respectively.Conclusion:The method is simple,accurate and reliability,and it can be used to provide scientific evidences for determination of Luteolin-7-O-β-D-Glucuronide,rosmarinus acid and tilianin in Mongolia D.moldavica Extracts.
