摘要
目的探讨云香水仙WD40基因在花色形成中的作用。方法基于前期的转录组数据,以云香水仙为材料,采用RT-PCR技术对WD40基因进行克隆,并结合Real-time PCR检测其在不同花期的表达水平。结果克隆得到1个WD40基因,命名为NtWD40,开放阅读框长为1020 bp,共编码339个氨基酸。同源性分析表明,云香水仙与拟南芥、苹果、矮牵牛及苜蓿的相似系数分别为68%、68%、68%和69%。Real-time PCR分析表明:始花期NtWD40的表达量较其他花期有明显差异。结论NtWD40可能参与云香水仙类黄酮途径相关色素的合成。
Objective To explore the role of WD40 gene in flower color formation of Yunxiang Narcissus tazetta L..Method Based on former transcriptome data,a WD40 gene named NtWD40 was cloned by RT-PCR with Yunxiang Narcissus tazetta L as the material,and its expression level at different flowering stages was detected by real-time PC.Result A WD40 gene,named ntwd40,was cloned and its open reading frame was 1020 BP long,encoding 339 amino acids.The amino acid sequence homology analysis showed that the similarity coefficients of Yunxiang Narcissus tazetta L.with Arabidopsis,apple,Petunia and alfalfa were 68%,68%,68%and 69%,respectively.Real-time PCR results further showed that the relative expression of NtWD40 gene at the initial flowering stage was significantly different from other flowering stages.Conclusion NtWD40 might be essential in the pigment synthesis correlated to the flavonoid pathway of Yunxiang Narcissus tazetta L..
