灰树花多糖对胃溃疡大鼠肠激素及氧化因子的影响 被引量:1

Effects of Griflola Folsa Polysaccharide on Intestinal Hormones and Oxidative Factors in Rats with Gastric Ulcer

摘要 探讨灰树花多糖对胃溃疡大鼠胃泌素(gastrin,GAS)、生长抑素(somatostatin,SS)、胃动素(motilin,MTL)、超氧化物歧化酶(Superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)和谷胱甘肽过氧化物酶(Glutathione peroxidase,GSH-Px)含量的影响。选取SPF级健康SD雄性大鼠70只,随机分为空白组10只、实验组60只,实验组采用“乙酸灌胃法”制作大鼠胃溃疡模型,1周后将造模成功的50只大鼠随机分为模型组、奥美拉唑组和灰树花多糖低、中、高剂量组,每组各10只。奥美拉唑组灌胃125 mg/(kg·d)、灰树花多糖组大鼠灌胃低剂量0.243 g/(kg·d)、中剂量0.486 g/(kg·d)、高剂量0.972 g/(kg·d),1次/d,连续8周,观察各组大鼠状态,记录体质量和进食量、溃疡面积、溃疡指数;采用ELISA法检测大鼠血清GAS(pg/mL)、SS(pg/mL)、MTL(pg/mL)水平;采用胺法测定SOD的活力、 TBA比色法测定MDA、 GSH-Px的含量。模型组大鼠较空白对照组GAS、SS、MDA升高(P<0.05或P<0.01),MTL、SOD、GSH-Px降低(P<0.05或P<0.01),药物干涉组随着灰树花多糖浓度的提高SOD、GSH-Px升高(P<0.05或P<0.01),GAS、MTL、SS、MDA指标降低(P<0.05或P<0.01),灰树花多糖具有降低胃肠激素分泌以及提高抗氧化酶(SOD、GSH-Px)含量的作用。 To investigate the effects of Griflola frondosa polysaccharide on the content of gastrin(GAS),somatostatin(SS),motilin(MTL),superoxide dismutase(SOD),malondialdehyde(MDA) and glutathione peroxidase(GSH-Px) in rats with gastric ulcer.70 SPF healthy SD male rats were selected and randomly divided into a blank group(10) and an experimental group(60),The experimental group used "acetic acid gavage" to make rat gastric ulcer models.After one week,50 rats with successful modeling were randomly divided into model group,omeprazole group,and low-dose,medium-dose and high-dose groups of foldash polysaccharide,with 10 rats in each group.Omeprazole group 125 mg/(kg·d) and griflola folsa polysaccharide group were given intragastric administration at low dose 0.243g/(kg·d),medium dose 0.486g/(kg·d) and high dose 0.972g/(kg·d),once a day for 8 weeks,to observe the status of rats in each group and record body weight,food intake,ulcer area and ulcer index;The serum levels of gas(pg/mL),SS(pg/mL) and MTL(pg/mL) were detected by ELISA;The activity of SOD was determined by amine method,and the contents of MDA and GSH PX were determined by TBA colorimetry.Compared with blank control group,GAS,SS and MDA in model group increased(P<0.05 or P<0.01),while MTL,SOD and GSH-Px decreased(P<0.05 or P<0.01).With the increase of the concentration of Grifola frondosa polysaccharide,the SOD,GSH PX increased(P<0.05 or P<0.01),and the gas,MTL,SS,MDA indexes decreased(P<0.05 or P<0.01) in the drug intervention group.Grifola frondosa polysaccharide can reduce gastrointestinal hormone secretion and increase the content of antioxidant enzymes(SOD,GSH PX).
