小鼠灌肠装置的改进与应用

摘要 目的改进小鼠灌肠实验装置,优化小鼠灌肠实验方法,解决传统灌肠方法的不足。方法使用6Fr导尿管联合自制肛塞改进小鼠灌肠装置,优化小鼠灌肠方法,通过小鼠灌肠实验,与传统灌肠方法进行比较。将60只雌性昆明小鼠随机分为5组(n=12),空白组不做处理,A、B、C组分别使用传统方法中的聚乙烯管、直头灌胃针、6Fr导尿管进行实验,D组使用改良后的灌肠装置6Fr导尿管联合自制肛塞进行实验,每组均给予0.4ml生理盐水进行灌肠,每日1次,连续14d。记录对比各组小鼠的一般状态及生存率;观察灌肠后漏液情况并进行评分;收集灌肠14d后小鼠的新鲜粪便并综合粪便性状和隐血或血便情况进行粪便评分;实验结束后,剖取肠组织,通过HE染色观察肠组织病理损伤情况。结果C、D组小鼠一般状态稍好于A、B组,各组间小鼠的存活率均无统计学差异(P>0.05);D组灌肠后漏液评分低于A、B组(P<0.05);D组粪便评分低于A组(P<0.05);HE染色显示,D组结肠组织病理损伤情况相较于A、B组明显减轻。结论改良后的小鼠灌肠装置比传统灌肠方法减少了灌肠后漏液,减轻了结肠及肛周组织损伤,具有操作简单、材料廉价的优点,可为相关研究提供借鉴。 Objective To improve the experimental apparatus,optimize the experimental method of mice enema and solve the shortcomings of traditional enema methods.Methods The 6Fr catheter combined with self-made anal plug was used to improve the mice enema device and optimize the mice enema method.The mice enema experiment was conducted to compare with the traditional enema methods.60 female Kunming strain mice were randomly divided into 5 groups(n=12).No treatment was given to blank group,the traditional apparatus polyethylene tube,straight head gavage needle and 6Fr catheter were used in group A,B and C respectively,the improved enema device,6Fr catheter combined with self-made anal plug was used in group D for the experiment.Each group was given 0.4ml normal saline for enema,once a day,for 14 consecutive days.The general state and survival rate of mice in each group were recorded and compared.The fluid leakage after enema was observed and scored.Fresh feces of mice 14 days after enema were collected and scored by combining feces characteristics and occult blood or bloody stool.After the experiment,intestinal tissue was taken out and pathological damage of intestinal tissue was observed by HE staining.Results The comparison of the general state of mice in each group showed that the groups C and D was slightly better than that in groups A and B,and survival analysis showed that there was no significant difference in the survival rate of mice in each group(P>0.05).The scores of post-enema leakage showed that the group D was significantly lower than that in groups A and B(P<0.05);Fecal scores of group D was significantly lower than that in group A(P<0.05);HE staining showed that the pathological injury of colonic tissue in group D was significantly reduced compared with that in groups A and B.Conclusion Compared with the traditional enema methods,the modified mice enema device can reduce the leakage after enema and reduce the damage of colon and perianal tissue.It is easy to operate and cheap,which can provide reference for related research.
