摘要
为N-杂环卡宾金属配合物在药物化学中的应用提供新的思路,合成了1-丁基-3-(2-甲基-8-丁氧基喹啉)咪唑六氟磷酸盐(L)和1-苄基-3-(2-甲基-8-丁氧基喹啉)咪唑六氟磷酸盐(M),并与金属阳离子配位反应,得到新型N-杂环卡宾金属配合物(L-Hg、M-Hg和M-Ag),其结构经^(1)H NMR,^(13)C NMR和X-单晶衍射表征。单晶结构结果分析表明:配合物L-Hg为单斜晶系、C 2/c空间群,汞原子与2个卡宾碳原子配位形成直线结构;配合物M-Hg为三斜晶系、P-空间群,汞原子与2个配体卡宾碳原子形成二配位体结构;配合物M-Ag为单斜晶系、P2(1)/c空间群,银原子分别与卡宾碳、氧原子、喹哪啶氮原子以及银原子配位,形成少见的四配位双核银卡宾化合物。通过最低抑菌浓度法发现M-Ag的抑菌浓度约为0.1 mg·mL^(-1)。
It provides a new idea for the application of N-heterocyclic carbene metal complexes in chemical Medicinal chemistry.In this paper,1-butyl-3-(2-methyl-8-butoxyquinoline)imidazole Hexafluorophosphate(L)and 1-benzyl-3-(2-methyl-8-butoxyquinoline)imidazole Hexafluorophosphate(M)were synthesized and coordinated with metal cations to obtain new N-heterocyclic carbene metal complexes(L-Hg,M-Hg and M-Ag),All the new compounds were characterized by^(1)H NMR,^(13)C NMR and X-ray single crystal diffraction.The single crystal X-ray diffraction analysis revealed that the complex L-Hg was monoclinic systemand the C 2/c space group,which was a straight line structure consisting of a mercury atom coordinated with two carbine carbon atoms.The structure of complex M-Hg was the triclinic system and the P-space group,which was taken a binary ligand structure between mercury atom and two carbine carbon atom.The complex M-Ag was the monoclinic system and crystallized in the P 2(1)/c space group,containing a rare argentum-argentum bond,the bonding argentum with carbine carbon atom and argentum atom coordination with oxygen atom and nitrogen atom on quinaldine,which was a rare tetracoordinated dinuclear silver carbene complex.The minimum inhibitory concentration method revealed that the inhibitory concentration of M-Ag was around 0.1 mg·mL^(-1).
