一种新型黏度和硫化氢荧光探针的制备及应用

摘要 黏度是一个重要的微环境参数,同时硫化氢(H_(2)S)也是一种重要的气体分子,这两者都会参与和影响许多生理和病理过程.因此,对细胞内黏度和H_(2)S浓度变化的检测具有实际意义.设计合成了荧光探针Mito-ANP,实现了同时检测体系内黏度和H_(2)S.当体系的黏度增加时,探针Mito-ANP的旋转被禁止,从而释放出红色的荧光.而加入H_(2)S后,探针Mito-ANP的双键与H_(2)S发生亲核加成反应释放出绿色荧光.探针Mito-ANP与硫化氢(H_(2)S)和黏度呈良好的线性关系,并且对H_(2)S具有专一的选择性.此外,探针Mito-ANP还能成功地用于神经细胞中硫化氢和黏度变化的荧光成像. Viscosity is an essential micro-environmental parameter and hydrogen sulfide(H_(2)S)is a critical gaseous signaling molecule,which are both related to various physiological and pathological processes.It has practical significance to detect the concentration of H_(2)S and viscosity change.In this work,we developed a fluorescent probe Mito-ANP for simultaneous detecting H_(2)S and viscosity.After the increase of viscosity,the rotation of the probe is prohibited and an intense red fluorescence is released.While,upon the addition of H_(2)S,the C=C of the probe can react with H_(2)S via the nucleophilic addition to form Mito-ANP-H_(2)S and a strong green fluorescence can be observed.Moreover,the probe Mito-ANP exhibited excellent relationship between viscosity and H_(2)S.In addition,the probe Mito-ANP was successfully to used for fluorescent imaging of viscosity and H_(2)S changes in neurocyte cells.
