摘要
【目的】探明影响木姜叶柯[Lithocarpus litseifolius(Hance)Chun.]老叶根皮苷提取的主要因素及其最适提取工艺,为开发利用木姜叶柯产品提供科学依据。【方法】以贵州黔西南州百年木姜叶柯树的老叶为材料,采用超声波提取方法,结合单因素和正交试验研究不同提取溶剂(70%甲醇、70%乙醇、0.5%氢氧化钠和水)、体积分数(50%、60%、70%、80%和90%)、超声波功率(30 W、50 W、70 W、90 W和110 W)、提取温度(30℃、40℃、50℃、60℃、70℃和80℃)、提取时间(10 min、20 min、30 min、40 min、50 min和60 min)及料液比(1∶10、1∶20、1∶30、1∶40、1∶50和1∶60,g/mL)等因素对木姜叶柯根皮苷提取的影响。【结果】料液比对提取率的影响最大(1.93%),其次是温度(1.19%)、时间(0.57%)和超声波功率(0.02%);根皮苷的最适提取条件为提取溶剂70%甲醇、料液比1∶20、提取时间50 min、提取温度70℃、功率90 W,在此条件下根皮苷提取率为3.81%。【结论】木姜叶柯老叶根皮苷的最适提取工艺参数为料液比(g/mL)1∶20,提取温度70℃,提取时间50 min,提取功率90 W。
【Objective】The main factors affecting the extraction of phloridzin from the old leaves of Lithocarpus litseifolius and the optimum extraction process were investigated,which provided the scientific basis for developing and exploiting L.litseifolius products.【Method】With the old leaves on 100-year-old L.litseifolius in Qianxinan,Guizhou as the material,ultrasonic extraction method,single factor and orthogonal test were used to study different extraction solvents(70%methanol,70%ethanol,0.5%NaOH and water),volume fraction(50%,60%,70%,80%and 90%),ultrasonic power(30 W,50 W,70 W,90 W and 110 W),the extraction temperature(30℃,40℃,50℃,60℃,70℃and 80℃),the extraction time(10 min,20 min,30 min,40 min,50 min and 60 min)and material-liquid ratio(1∶10,1∶20,1∶30,1∶40,1∶50 and 1∶60),on the extraction effect of phloridzin were analyzed.【Result】The material-liquid ratio had the greatest influence on extraction rate(1.93%),followed by temperature(1.19%),extraction time(0.57%)and ultrasonic power(0.02%).The optimal extraction conditions were as follows:70%methanol,material-liquid ratio with 1∶20,extraction time with 50 min,extraction temperature with 70℃and ultrasonic power with 90 W.The extraction rate of phloridzin under this condition was 3.81%.【Conclusion】The extraction process of phloridzin from L.litseifolius old leaves were optimized,and the optimal extraction process included 1∶20 g/mL(material-liquid ratio),70℃(extraction temperature),50 min(extraction time),90 W(ultrasonic power).
