摘要
以品种里约作为试验材料,分别以矾根的根茎基部幼叶、叶柄和根为外植体,筛选最佳研究不同消毒处理方式和植物生长调节剂对其不定芽诱导、再生和生根的影响情况,建立其组培快繁体系。结果显示,适宜矾根里约初代诱导培养基为MS+0.2 mg/L 6-BA+0.1 mg/L NAA;继代培养基为MS+0.2 mg/L 6-BA+0.1 mg/L IAA;生根培养基为1/2 MS+1.0 mg/L IBA,生根率95.08%;生根苗移栽驯化基质为泥炭和珍珠岩3∶1的混合基质,成活率达82.91%。
The young leaves,petioles and roots of Heuchera micrantha Rio were selected as explants,and the effects of different disinfection treatments and plant growth regulators on adventitious bud induction,regeneration and rooting were studied to establish a tissue culture and rapid propagation system.The results showed that MS+0.2 mg/L 6-BA+0.1 mg/L NAA was the appropriate medium for the initial induction of Heuchera micrantha.The secondary medium was MS+0.2 mg/L 6-BA+0.1 mg/L IAA.The rooting medium was 1/2 MS+1.0 mg/L IBA,and the rooting rate was 95.08%.The root seedlings were transplanted and acclimated with peat and perlite 3∶1,the survival rate was 82.91%.
