复方滇鸡血藤颗粒的指纹图谱建立及4个木脂素类成分含量测定

Establishment of fingerprint of Compound Dianjixueteng Granule and determination of four lignans

目的:建立复方滇鸡血藤颗粒的指纹图谱,并建立颗粒中4个木脂素类成分同时测定的高效液相色谱(HPLC)方法,为其质量控制提供参考。方法:采用HPLC法建立复方滇鸡血藤颗粒的指纹图谱,以对照品进行共有峰指认,通过制剂与各单味药材的相关性分析进行共有峰归属,通过相似度评价结合化学模式识别分析11批样品的质量差异,同时以4个木脂素类成分戈米辛D、内南五味子素、异型南五味子丁素、南五味子素为指标性成分,建立其含量测定方法。结果:建立了复方滇鸡血藤颗粒的11批样品指纹图谱,共标定20个共有峰,其中1号峰为马钱苷、3号峰为羟基红花黄色素A、4号峰为杯苋甾酮、6号峰为大豆苷元、13号峰为戈米辛D、17号峰为内南五味子素、19号峰为异型南五味子丁素、20号峰为南五味子素;各批次样品间相似度均大于0.97,可聚为2类,4个主成分的累积贡献率达89.764%;4个木脂素类成分戈米辛D、内南五味子素、异型南五味子丁素、南五味子素的含量分别为386.35~426.96、280.69~307.66、693.31~740.91、188.66~215.59μg·g^(-1)。结论:所建立的指纹图谱及含量测定方法可为复方滇鸡血藤颗粒的质量控制与评价提供依据。

OBJECTIVE To establish the fingerprint of Compound Dianjixueteng Granule and establish an HPLC method for simultaneous determination of four lignans in the granule,providing reference for their quality control.METHODS The fingerprint of Compound Dianjixueteng Granule was established by HPLC method,the common peaks were identified with the reference substances,and the original attribute of the common peaks were confirmed by the correlation analysis between the preparation and each single herb.The quality difference of 11 batches of samples was analyzed by similarity evaluation and chemical pattern recognition.At the same time,the content determination method was established by taking gomisin D,interiorin,heteroclitin D and kadsurin as indicative components.RESULTS The fingerprints of 11 batches of Compound Dianjixueteng Granule was established,and 20 common peaks were calibrated,among which,peak 1 was loganin,peak 3 was hydroxysafflor yellow A,peak 4 was cyasterone,peak 6 was daidzein,peak 13 was gomisin D,peak 17 was interiorin,peak 19 was heteroclitin D and peak 20 was kadsurin.The similarity between the samples of each batch was>0.97,which could be grouped into two categories.The cumulative contribution rate of the four main components was 89.764%.The content of four lignans,gomisin D,interiorin,heteroclitin D and kadsurin,were 386.35-426.96,280.69-307.66,693.31-740.91,188.66-215.59μg·g^(-1),respectively.CONCLUSION The established fingerprint and content determination method can provide basis for the quality control and evaluation of Compound Dianjixueteng Granule.
