鸡Myf5蛋白结构与功能的生物信息学分析

摘要 [目的]分析并预测鸡Myf5蛋白的结构与生物学功能。[方法]采用ProtParam、ProtScale、SingnalP-4.1、STRING等软件对鸡Myf5蛋白的序列结构及互作蛋白进行分析。[结果]该蛋白由258个氨基酸构成,分子式为C_(1221)H_(1915)N_(359)O_(391)S_(18),氨基酸残基中带负电荷的(Asp+Glu)共有34个,氨基酸残基中带正电荷的(Arg+Lys)共有29个;理论等电点(pI)为5.86,半衰期为30 h,不稳定性指数为97.59(>40);亲水性平均值(GRAVY)为-0.701,脂溶指数为54.92;其二级结构主要以无规则卷曲为主,占比63.57%,其次为α-螺旋,占比29.84%;该蛋白定位于细胞核中,不具备跨膜性,共有74个潜在的磷酸化位点,无信号肽,为亲水性不稳定蛋白。蛋白互作分析结果显示,鸡Myf5关联的蛋白有PAX7、PAX3、SOX10、MYOD1、MYOG和SDC4,进化分析结果显示不同物种间该蛋白序列具有较高的保守性。[结论]该蛋白可能在鸡肌肉的生长发育过程中发挥重要作用,为进一步探索其功能奠定基础。 [Objective]The study aimed to analyze and predict the structure and biological function of chicken Myf5.[Method]The structure of sequence and interacting protein of chicken Myf5 were analyzed by ProtParam,ProtScale,SingNALP-4.1,STRING and other softwares.[Result]The protein was composed of 258 amino acids with the molecular formula of C_(1221)H_(1915)N_(359)O_(391)S_(18).There were 34 negatively charged amino acid residues(Asp+Glu)and 29 positively charged amino acid residues(Arg+Lys).The theoretical isoelectric point(pI)was 5.86,the half-life was 30 h,and the instability index was 97.59(>40).The average value of GRAVY was-0.701 and the lipid solubility index was 54.92.The secondary structure of this protein is mainly random coil,accounting for 63.57%,followed byα-helix,accounting for 29.84%.The protein is localized in the nucleus,whicn has 74 potential phosphorylation sites without transmembrane property and signal peptide.It is a hydrophilic unstable protein.Protein interaction analysis showed that chicken Myf5 was associated with PAX7,PAX3,SOX10,MYOD1,MYOG and SDC4.Evolutionary analysis showed that the sequence of this protein was highly conserved among different species.[Conclusion]This protein may play an important role in the growth and development of chicken muscle,laying a foundation for further exploration of its function.
