摘要
目的筛选炎症性肠病患者血清外泌体中的差异蛋白,分析其功能及生物学过程。方法选取2021年12月至2022年3月在郑州大学第一附属医院住院确诊的炎症性肠病患者(炎症性肠病组)与健康体检者(健康对照组),各3例,收集患者血清样本,采用外泌体提取试剂盒提取血清外泌体,采用4D-Label-free定量蛋白质组学技术对两者血清中的外泌体蛋白进行鉴定和定量分析,筛选出差异蛋白,通过生物信息学分析差异蛋白的来源及生物学功能。结果共鉴定到5765个肽段,891个蛋白,以差异倍数>2且P<0.05为标准共筛选出55种差异蛋白,与健康对照组相比,炎症性肠病组上调蛋白38种,下调蛋白17种。生物信息学分析这些差异蛋白主要集中在细胞内过程、刺激应答、生物学调节、代谢过程等生物学过程,与神经变性、细菌及病毒感染相关,还参与了Ras信号、钙信号、cCMP-PKG信号等信号通路。结论4D-Label-free定量蛋白质组学技术筛选出的差异蛋白可能作为炎症性肠病早期诊断的血清学标志物及治疗靶点。
Objective To screen differential proteins in serum exosomes of patients with inflammatory bowel disease(IBD),and analyze their functions and biological processes.Methods Serum samples were collected from 3 IBD patients(IBD group)and 3 healthy people(healthy control group)who were hospitalized and diagnosed in the First Affiliated Hospital of Zhengzhou University from Dec.2021 to Mar.2022.Serum exosomes were extracted with an exosome extraction kit.The exosomal proteins in the serum from the two groups were identified and quantitatively analyzed by 4D-Label-free quantitative proteomics technology.The source and biological function of the differential proteins were analyzed by bioinformatics.Results A total of 5765 peptide segments and 891 proteins were identified.55 differentially expressed proteins were screened based on the standard of difference multiple>2 and P<0.05.Compared with healthy controls,38 proteins were up-regulated and 17 proteins were down-regulated in IBD group.Bioinformatics analysis results showed that these differential proteins were mainly concentrated in biological processes such as cellular process,response to stimulus,biological regulation,metabolic process,which were related to neurodegeneration,bacterial and viral infections,and also participates in signal pathways such as Ras signal,calcium signal and cCMP-PKG signal.Conclusion The differential proteins screened by 4D-Label-free quantitative proteomics may be used as serological markers and therapeutic targets for early diagnosis of IBD.
