基于网络药理学探讨桑叶-菊花治疗小儿抽动障碍的作用机制 被引量:2

摘要 目的基于网络药理学方法分析桑叶-菊花在预防小儿抽动障碍方面的药理机制,为临床上治疗小儿抽动障碍提供参考和依据。方法通过中药系统药理学分析平台(TCMSP)数据库获取桑叶-菊花药对成分的主要活性化学成分及其相关作用靶点;通过GeneCards、OMIM、Drugbank、Pharmgkb及TTD数据库获取小儿抽动障碍相关靶点,利用String平台进行蛋白质-蛋白质相互作用(Protein-Protein Interaction,PPI)分析,构建PPI网络并挖掘网络中潜在的蛋白质功能模块。在Cytoscape 3.9.1进行“药物-成分-靶点”分析,对其参与的生物过程及通路进行富集并将结果进行可视化,构建“桑叶菊花药对-小儿抽动障碍靶点-通路”网络。结果桑叶-菊花治疗TD的核心活性成分为槲皮素、β-谷甾醇、山柰酚、木犀草素等。核心靶点有TNF、IL-6、FOS、AKT1等。桑叶-菊花调治小儿抽动障碍的生物学通路主要作用于MAPK信号通道、肿瘤坏死因子信号通路、T细胞受体信号等通路。结论本研究初步揭示了桑叶-菊花可多成分、多靶点、多通路治疗小儿抽动障碍的作用机制,为临床用药提供依据。 OBJECTIVE To analyze the active components,targets and key pathways of Sangye-Juhua in regulating tic disorders based on network pharmacology,and provide reference and basis for the clinical application and promotion of traditional Chinese medicine.METHODS The main active chemical components of Sangye-Juhua and their related targets were obtained from the TCMSP database.The targets related to tic disorders were obtained from GeneCards,OMIM,TTD,and GKB databases,and the String platform performed PPI analysis,constructed a PPI network and excavated potential protein functional modules in the network.The Cytoscape 3.9.1 software was used to analyze the“drug-component-target”,the biological processes and pathways were enriched and the results were visualized and to construct the“Sangye-Juhua-tic disorders target-pathway”network.RESULTS The core active ingredients of Sangye-Juhua for the treatment of tic disorders were quercetin,β-Sitosterol,kaempferol,luteolin,etc.The core targets were TNF、IL-6、FOS、AKT1,etc.The biological pathway of Sangye-Juhua for treating tic disorders were mainly acted on MAPK signaling pathway,TNF signaling pathway,T cell receptor signaling pathway,etc.CONCLUSION This study has initially revealed the multi-component,multi-target,and multi-path mechanism of Sangye-Juhua on tic disorders,which provides a basis for the clinical medication.
