SCF/c-kit信号通路对乳腺癌侵袭转移调控的作用及机制研究

摘要 目的:探讨SCF/c-kit信号通路对乳腺癌侵袭转移调控的作用及机制研究。方法:采用人乳腺癌细胞株MCF-7构建体外模型。将细胞分为空白对照组、SCF过表达组和SCF抑制组。采用MTT法检测细胞24h、48h、72h的增殖率,采用Transwell实验检测细胞24h、48h、72h的侵袭情况。结果:SCF过表达组在24h、48h、72h的细胞增殖率均显著大于SCF抑制组和空白对照组(P<0.05),且SCF抑制组细胞增殖率显著低于空白对照组(P<0.05);SCF过表达组在24h、48h、72h的细胞侵袭数均显著大于SCF抑制组和空白对照组(P<0.05),且SCF抑制组细胞侵袭数显著低于空白对照组(P<0.05)。结论:SCF/c-kit信号通路的激活可增强乳腺癌细胞的增殖和侵袭能力,靶向抑制SCF/c-kit信号通路的表达可有效逆转对乳腺癌细胞增殖和侵袭的促进作用。 Objective:To investigate the effect and mechanism of SCF/c-kit signaling pathway on invasion and metastasis of breast cancer.Methods:Human breast cancer cell line MCF-7 was used to construct the model in vitro.The cells were divided into blank control group,SCF overexpression group and SCF inhibition group.MTT assay was used to detect cell proliferation rate at 24h,48 h and 72 h,and Transwell assay was used to detect cell invasion at 24h,48h and 72h.Results:The cell addition rate of SCF overexpression group at 24h,48h and 72h was significantly higher than that of SCF inhibition group and blank control group(P<0.05),and the cell addition rate of SCF inhibition group was significantly lower than that of blank control group(P<0.05).The number of cell invasion in SCF overexpression group at 24h,48h and 72h was significantly higher than that in SCF inhibition group and blank control group(P<0.05),and the number of cell invasion in SCF inhibition group was significantly lower than that in blank control group(P<0.05).Conclusion:Activation of SCF/c-kit signaling pathway can enhance the proliferation and invasion ability of breast cancer cells,and targeted inhibition of SCF/c-kit signaling pathway expression can effectively reverse the promoting effect on the proliferation and invasion of breast cancer cells.
