佛手散改善谷氨酸钠诱导的PC12 细胞氧化应激损伤的作用机制

摘要 目的探讨佛手散改善谷氨酸钠诱导的PC12细胞氧化应激损伤的作用机制。方法将PC12细胞随机分为对照组、模型组、佛手散高剂量组、佛手散中剂量组、佛手散低剂量组。分别给予佛手散低剂量组、佛手散中剂量组、佛手散高剂量组32 mg/mL、64 mg/mL和128 mg/mL佛手散处理,给予对照组和模型组PBS处理。除对照组外,对其余4组PC12细胞建立谷氨酸钠诱导的氧化应激损伤模型。比较各组PC12细胞的超氧化物歧化酶(SOD)、谷胱甘肽、丙二醛和活性氧簇的含量,以及凋亡相关蛋白[Caspase-3、B细胞淋巴瘤2(Bcl-2)和Bcl-2相关X蛋白(Bax)]的表达水平。结果模型组PC12细胞的SOD和谷胱甘肽含量、Bcl-2蛋白表达水平均低于对照组,丙二醛和活性氧簇含量、Caspase-3和Bax蛋白表达水平均高于对照组(均P<0.05)。佛手散低剂量组、佛手散中剂量组、佛手散高剂量组PC12细胞的丙二醛和活性氧簇含量、Caspase-3和Bax蛋白表达水平均低于模型组,SOD和谷胱甘肽含量、Bcl-2蛋白表达水平均高于模型组(均P<0.05),且上述指标的变化总体上呈剂量依赖性。结论佛手散可以通过提高细胞的抗氧化能力及减少细胞凋亡,来抑制谷氨酸钠诱导的PC12细胞氧化应激损伤,从而发挥保护神经的作用。 Objective To explore the mechanism of Foshousan for the amelioration of PC12 cells oxidative stress injury induced by sodium glutamate.Methods PC12 cells were randomly divided into control group,model group,high-dose Foshousan group,medium-dose Foshousan group,or low-dose Foshousan group.Foshousan with 32 mg/mL,64 mg/mL,and 128 mg/mL were given to the low-,medium-,and high-dose Foshousan groups for treatment,respectively,and the control and model groups received PBS treatment.Except for the control group,the oxidative stress injury model induced by sodium glutamate was established in PC12 cells of the remaining 4 groups.The contents of superoxide dismutase(SOD),glutathione,malondialdehyde,and reactive oxygen species,as well as expressions of apoptosis-related proteins with respect to Caspase-3,B-cell lymphoma 2(Bcl-2),and Bcl-2 associated X protein(Bax)were compared between PC12 cells of various groups.Results The contents of SOD,glutathione,and Bcl-2 protein expression of PC12 cells in the model group were lower than those in the control group,the contents of malondialdehyde,reactive oxygen species,and protein expressions of Caspase-3 and Bax of PC12 cells in the model group were higher than those in the control group(all P<0.05).The contents of malondialdehyde,reactive oxygen species,and protein expressions of Caspase-3 and Bax of PC12 cells in the low-,medium-,and high-dose Foshousan groups were lower than those in the model group,and the contents of SOD,glutathione,and Bcl-2 protein expression were higher in the low-,medium-,and high-dose Foshousan groups than in the model group(all P<0.05),and the indices as above was dose-dependent as a whole.Conclusion Foshousan can inhibit PC12 cells oxidative stress injury induced by sodium glutamate through increasing antioxidant capacity of cells,and decreasing apoptosis of cells,so as to exert protective effects for nerve.
