摘要
针对中熟桃品种黄妃存在大量种子败育,影响育种效率和进程的问题,春季以黄妃做母本,黄金蜜1号为父本进行杂交,8月初果实成熟后收集杂交种子,研究了不同基本培养基中加入不同植物生长调节剂与浓度的培养基上接种种胚的培养效果。结果表明,杂交种胚在基本培养基WPM上加入1 mg/L 6-BA+0.1 mg/L IBA的培养基上具有最高的发芽率;在同浓度IBA条件下,6-BA浓度越高,种胚的发芽率越高,幼苗苗高越高而发根数减少;IBA浓度与根的发育成正相关。
There is a large amount of seed abortion in the middle-maturing peach variety Huangfei,which greatly affects the efficiency and progress of breeding.In this study,Huangfei was used as the female parent in spring and three male parent materials were allocated to carry out the cross pollination experiment.The hybrids were collected after fruit ripening at the beginning of august,and the effects of different medium and different hormone concentrations on the germination of seed embryos were used.The results showed that during the selection of basic culture medium,hybrid embryos had the highest germination rate on WPM culture medium with 1 mg/L 6-BA+0.1 mg/L IBA;Under the same concentration of IBA,the higher the concentration of 6-BA,the higher the germination rate of seed embryo,the higher the plant height of the seedlings while the number of roots decreased;There is a positive correlation between IBA concentration and root development.By conducting this study,a suitable technical system for hybrid embryo culture of Huangfei was established.
