摘要
目的:建立检测鸭蛋中微量苏丹红Ⅳ的实验方法。方法:考察不同提取剂、展开剂对蛋黄液中苏丹红Ⅳ的薄层色谱分离效果,利用拉曼光谱法对分离斑点进行定性分析,验证薄层色谱分离效果;利用紫外可见分光光度法对分离后的斑点样品进行定量分析。结果:以石油醚-丙酮(2∶1)为提取剂,以正己烷-丙酮-乙酸乙酯(100∶6∶8)为薄层色谱展开剂,各组分分离效果好;以511 nm为测定波长,浓度与吸光度在0.50~25.00 mg·mL^(-1)范围内呈良好线性关系(R2=0.99957),检出限为0.07 mg·mL^(-1),回收率为99%~108%。结论:该薄层色谱-紫外可见分光光度法联用的方法操作简便,成本低,专属性强,适用于鸭蛋中微量苏丹红Ⅳ的测定。
Objective:To establish an experimental method for the determination of Sudan IV in duck eggs.Methods:The effect of different extractants and developing agents on the thin layer chromatographic separation of Sudan IV in egg yolk liquid was investigated.The separation spots were qualitatively analyzed by Raman spectroscopy to verify the separation effect of thin layer chromatography;Ultraviolet visible spectrophotometry was used to quantitatively analyze the spot samples after separation.Results:With petroleum ether acetone(2∶1)as the extracting agent and n-hexane acetone ethyl acetate(100∶6∶8)as the developing agent,each component was well separated;With 511 nm as the measuring wavelength,the concentration and absorbance are 0.50~25.00 mg.Linear relationship within the range of mL^(-1)(R2=0.99957),detection limit 0.07 mg·mL^(-1),the recovery is 99%~108%.Conclusion:The method is simple,low cost and specific,and suitable for the determination of Sudan IV in duck eggs.
