东方草莓匍匐茎化学成分及其神经保护活性

摘要 目的研究东方草莓Fragaria orientalis Losinsk匍匐茎的化学成分及其神经活性。方法东方草莓匍匐茎80%乙醇提取物采用硅胶、ODS、Sephadex LH-20、半制备HPLC进行分离纯化,根据理化性质及波谱数据鉴定所得化合物的结构。采用6-OHDA诱导的PC12细胞评价其神经保护活性。结果从中分离得到18个化合物,分别鉴定为meliasendanin D(1)、双酚A(2)、阿魏酸十六烷基酯(3)、octadecyl ferulate(4)、eicosanyl ferulate(5)、piceine(6)、香草酸(7)、反式肉桂酸(8)、顺式肉桂酸(9)、虹彩内酯(10)、ethylβ-D-xylopyranoside(11)、β-谷甾醇(12)、(9Z,12Z,15Z)-9,12,15-十八碳三烯酸(13)、(9Z,12Z,15Z)-9,12,15-十八碳三烯酸甲酯(14)、(9Z,12Z,15Z)-9,12,15-十八碳三烯酸乙酯(15)、(9Z,12Z)-9,12-十八碳二烯酸(16)、(9Z,12Z)-9,12-十八碳二烯酸甲酯(17)、(9Z,12Z)-9,12-十八碳二烯酸乙酯(18)。化合物1、5~8、10、11能升高6-OHDA诱导的PC12细胞存活率,其中化合物6、10最为显著,分别升高了11.8%、12.9%。结论化合物1~11、13~18均为首次从该植物中分离得到。化合物1、5~8、10、11具有神经保护活性。 AIM To study the chemical constituents from the stolons of Fragaria orientalis Lozinsk and their neuroprotective activities.METHODS The 80%ethanol extract from the stolons of F.orientalis was isolated and purified by silica gel,ODS,Sephadex LH-20 and semi-preparative HPLC,then the structures of obtained compounds were identified by physicochemical properties and spectral data.Their neuroprotective activities were evaluated by 6-OHDA-induced impaired PC12 cells.RESULTS Eighteen compounds were isolated and identified as meliasendanin D(1),bisphenol A(2),ferulic hexadecyl ester(3),octadecyl ferulate(4),eicosanyl ferulate(5),piceine(6),vanillic acid(7),trans-cinnamic acid(8),cis-cinnamic acid(9),iridolactone(10),ethylβ-D-xylopyranoside(11),β-sitosterol(12),(9Z,12Z,15Z)-9,12,15-octadecatrienoic acid(13),(9Z,12Z,15Z)-9,12,15-octadecatrienoic acid methyl ester(14),(9Z,12Z,15Z)-9,12,15-octadecatrienoic acid ethyl ester(15),(9Z,12Z)-9,12-octadecadienoic acid(16),(9Z,12Z)-9,12-octadecadienoic acid methyl ester(17),(9Z,12Z)-9,12-octadecadienoic acid ethyl ester(18).Compounds 1,5-8,10,11 could increase the survival rates of PC12 cells induced by 6-OHDA.Compounds 6,10 had the most significant effects,increasing by 11.8%and 12.9%,respectively.CONCLUSION Compounds 1-11,13-18 are isolated from this plant for the first time.Compounds 1,5-8,10,11 show neuroprotective activities.
