摘要
目的:构建20批黔产绿茶的HPLC指纹图谱,为科学评价和控制黔产绿茶的质量控制提供参考依据。方法:采用HPLC法,以Diamonsil C18(250×4.6 mm,5μm)为色谱柱,流动相以甲醇(A)-乙腈(B)-0.1%磷酸水溶液(C)进行梯度洗脱,流速为1.0 mL/min,检测波长210 nm,柱温35℃,进样量10μL。通过相似度分析(similarity analysis,SA)、聚类分析(cluster analysis,CA)、主成分分析(principal componentanalysis,PCA)对指纹图谱分析。结果:建立的20批黔产绿茶指纹图谱,标定15个共有峰,并指认了其中5个共有峰,相似度计算结果在0.610~0.966之间,聚类分析、主成分分析结果较为一致,均将样品分为3大类。SA、CA和PCA三种分析方法结果相互补充、相互映证,揭示贵州各产地绿茶具有一定相似性但也存在差异性。结论:本文建立的绿茶HPLC指纹图谱测定方法专属性较强、重复性好,该研究对贵州不同产地绿茶的鉴别提供了参考,可用于黔产绿茶的质量控制和品质评价。
Objective:To establish the fingerprint of 20 batches of green tea from Guizhou based on high performance liquid chromatography(HPLC),in order to provide a theoretical basis for improving the quality control of green tea.Methods:Based on the HPLC method,a Diamonsil C18 column(250×4.6 mm,5μm)was used with a gradient elution of methanol(A)-acetonitrile(B)-0.1%phosphoric acid aqueous solution(C),the flow rate was 1.0 mL·min-1,detection wavelength was 210 nm,column temperature was 35℃,injection volume was 10μL.The National Pharmacopoeia Commission recommended“Chinese medicine chromatographic fingerprint similarity evaluation system 2004 A version”was applied to establish a common model of green tea fingerprints,and adopt similarity analysis(SA),cluster analysis(CA)and principal component analysis(PCA)to evaluate 20 batches of green tea fingerprints from different origins in Guizhou.Results:20 batches of green tea fingerprints had 15 common peaks,5 common peaks were identified as epigallocatechin,catechin,epicatechin,gallocatechin gallate and epigallocatechin gallate,respectively.The similarity was calculated between 0.610 and 0.966,and the clustering analysis divided the samples into four categories,principal component analysis extracted the first three principal components,of which the cumulative contribution was 86.724%.The quality of green tea in Tongren district,Meitan of Zunyi district,and Weining of Bijie is better in Guizhou province.SA,CA and PCA analysis methods complemented and confirmed each other,revealing that the chemical composition of green tea of each origin was similar but still was different to some extent.Conclusion:The HPLC fingerprint method for green tea established in this paper is highly specific,reproducible,simple and reliable,and can be used for quality control and evaluation of green tea in Guizhou.
