摘要
目的:采用高效液相色谱法建立关黄柏叶中黄柏酮的含量测定方法。方法:高效液相色谱法,WondaSil C18 Superb色谱柱(150 mm×4.6 mm,5μm),乙腈-0.1%醋酸水(V∶V=49∶51)为流动相,流速为1.0 mL/min,检测波长为215 nm,柱温为30℃。结果:黄柏酮在1.68~52.74μg/mL范围内,线性关系良好(R2=0.9999)。供试品溶液在24 h内稳定性良好,方法的精密度高,平均加样回收率为102.2%,RSD为2.27%。关黄柏叶中黄柏酮的含量为1.604 mg/g。结论:所建立的方法可用于测定关黄柏叶中黄柏酮的含量。
Objective:To establish a method for the determination of Obacunone in Phellodendron amurense Rupr.leaves by HPLC.Methods:High-performance liquid chromatography,Chromatographic column:WondaSil C18 Superb(150 mm×4.6 mm,5μm);Mobile phase:acetonitrile-0.1% acetic acid water(V∶V=49∶51);Velocity:1.0 mL/min.The detection wavelength was set at 215 nm and the column temperature was 30℃.Results:The linear relationship was good in the range of 1.68~52.74μg/mL(R2=0.9999).The test solution has good stability within 24 h,and the method has high precision.The average recovery was 102.2% and RSD was 2.27%.The content of Obacunon in the leaves was 1.604 mg/g.Conclusion:The established method can be used for the determination of Obacunon in Phellodendron amurense Rupr.Leaves.
