苦参碱对结肠癌HT29细胞增殖及凋亡的临床研究

摘要 目的:探讨苦参碱对结肠癌HT29细胞增殖及凋亡的影响,并分析其作用机制。方法:选取2020年1月至2021年3月间于中国医学科学院肿瘤医院山西医院行结肠癌治疗的100例患者的资料进行分析,其中运用mFOLFOX6方案治疗的50例纳入对照组,另50例在mFOLFOX6方案治疗的中后期联合使用苦参碱治疗的纳入观察组。运用Western blot法检测两组患者B细胞淋巴瘤-2抗凋亡蛋白(Bcl-2)、Bcl-2相关蛋白X(Bax)、半胱氨酸蛋白酶-3(Caspase-3)及表皮生长因子受体(EGFR)水平,以分析苦参碱对HT29细胞增殖、凋亡的影响;并比较两组患者癌胚抗原(CEA)、糖类抗原19-9(CA19-9)及血管内皮生长因子(VEGF)水平在治疗前后的变化,以分析治疗效果。结果:治疗后,观察组患者的Bcl-2、EGFR水平均低于对照组,Bax和Caspase-3水平均高于对照组,差异均具有统计学意义(P<0.05);观察组患者CEA、CA19-9及VEGF水平均低于对照组,差异均具有统计学意义(P<0.05)。结论:苦参碱具有一定的抗肿瘤作用,其可以抑制结肠癌HT29细胞的增殖,诱导其凋亡,并通过调节Bcl-2/Bax、EGFR和Caspase-3等相关信号通路来发挥其作用。 Objective Explore the effect of matrine on the proliferation and apoptosis of colon cancer HT29 cells,and analyze its mechanism of action.Methods 100 patients who underwent colon cancer treatment at Shanxi Hospital of Cancer Hospital Chinese Academy of Medical Sciences from January 2020 to March 2021 were selected for analysis.Among them,50 patients treated with the mFOLFOX6 regimen were included in the control group,while the other 50 patients treated with the combination of matrine in the middle and later stages of the mFOLFOX6 regimen were included in the observation group.Western blot was used to detect the levels of B-cell lymphoma-2(Bcl-2),Bcl-2-associated X protein(Bax),cysteine-aspartic acid protease-3(caspase-3),and epidermal growth factor receptor(EGFR)in Bcl-2 in the two groups,the effects of matrine on the proliferation and apoptosis of HT29 cells were analyzed;The levels of carcinoembryonic antigen(CEA),carbohydrate antigen 19-9(CA19-9)and vascular endothelial growth factor(VEGF)in the two groups were compared before and after treatment to analyze the therapeutic effect.Results After treatment,the levels of Bcl-2 and EGFR in the observation group were lower than those in control group,while the levels of Bax and Caspase-3 were higher than those in the control group,the differences were statistically significant(P<0.05).The levels of CEA,CA19-9 and VEGF in the observation group were lower than those in the control group,the differences were statistically significant(P<0.05).Conclusion Matrine has a certain anti-tumor effect,which can inhibit the proliferation of colon cancer HT29 cells,induce apoptosis,and exert its effects by regulating related signaling pathways such as Bcl-2/Bax,EGFR,and caspase-3.
