摘要
目的:观察端粒锌指相关蛋白(TZAP)在乳腺导管癌及癌旁非瘤组织中的表达及其临床意义。方法:采用免疫组化法检测145例乳腺浸润性导管癌、35例导管原位癌及癌旁正常乳腺组织中TZAP的表达,分析其表达与临床病理特征的关系。结果:与癌旁正常乳腺组织100%强阳性相比(180/180),TZAP蛋白在乳腺导管内癌中虽然都有着色(总阳性率100%,35/35),但总体表达下调(其中强阳性率5.71%,2/35;中等阳性率68.57%,24/35;弱阳性率25.71%,9/35);在浸润性导管癌中表达下调更明显(阳性率为45.52%,66/145;阴性率为54.48%,79/145)。TZAP阴性的病例具有组织学分级较高、肿瘤体积较大、有淋巴结转移、HER2基因有扩增、Ki67增殖指数较高、TNM分期较高的特点,表明TZAP表达下调与乳腺导管癌的发生和进展有关。结论:TZAP在乳腺导管癌组织中表达下调,可能在其发生和恶性进展中起重要作用。
Objective:To detect the expression and clinical significance of telomere zinc finger-associated protein(TZAP) in breast ductal carcinoma tissues and paired paraneoplastic noncancerous breast tissues.Methods:145 cases of invasive ductal carcinoma tissues,35 cases of breast intraductal carcinoma tissues and paired paraneoplastic noncancerous breast tissues were subjected to immunohistochemistry.The relationship between the expressions of TZAP and its clinicopathological parameters were analyzed.Results:Compared with paraneoplastic noncancerous breast tissues(100% strong stained,180/180),TZAP protein was stained in intraductal carcinoma(100% positive,35/35),but its overall expression was down-regulated(with strong positive rate of 5.71%,2/35;moderate positive rate of 68.57%,24/35;weak positive rate of 25.72%,9/35).TZAP protein was more significantly downregulated in invasive ductal carcinoma tissues(with positive rate of 45.52%,66/145;negative rate of 54.48%,79/145).TZAP negative cases were characterized by higher histological grade,larger tumor size,positive lymph node metastasis,amplification of HER2 gene,higher proliferation index of Ki67 and higher TNM clinical stage,indicating that the downregulation of TZAP protein was associated with the carcinogenesis and progression of breast ductal carcinoma.Conclusion:The downregulated TZAP in breast ductal carcinoma tissues may be involved in carcinogenesis and progression of breast ductal carcinoma.
