摘要
目的:探究微小染色体维持蛋白6(Mini chromosome maintenance complex component 6,MCM6)在结直肠癌(Colorectal cancer,CRC)中的表达情况,以及与临床病理特征、预后及肿瘤免疫浸润的相关性。方法:基于TCGA数据库比较MCM1~10在CRC及正常组织中的表达;采用Kaplan-Meier生存曲线等分析MCM1~10的表达与CRC诊断、预后的关系;利用UALCAN与HPA数据库验证MCM6的表达并探究其在CRC不同临床阶段的表达变化;采用Cox回归分析MCM6的表达与CRC预后及临床病理特征的相关性;利用GO和KEGG分析MCM6潜在功能;利用STRING数据库构建蛋白-蛋白互作网络,探索MCM6调控网络和核心基因;基于TIMER数据库分析MCM6与免疫浸润的相关性。结果:MCM2~8、MCM10在CRC组织中高表达,并对CRC患者预后的评估有诊断意义。其中,MCM6高表达的CRC患者预后显著提高;MCM6高表达与CRC的临床分期和临床病理特征也显著相关;GO与KEGG分析结果提示,MCM6可能参与细胞周期和RNA转运等生物进程;MCM6还与多种免疫细胞浸润水平显著相关。结论:MCM6在CRC中高表达,并与CRC发展、预后、临床病理特征及肿瘤免疫浸润密切相关。MCM6可成为CRC诊断、预后的生物标志物以及潜在的治疗靶点。
Objective:To explore the expression of mini chromosome maintenance complex component 6(MCM6)in colorectal cancer(CRC)and its correlation with clinicopathological features,prognosis,and tumor immune infiltration.Methods:The expression differences of MCM1-10 in CRC and normal tissues were compared based on the cancer genome atlas(TCGA)database.Kaplan-Meier survival curve was used to analyze the relationship between the expression of MCM1-10 and the diagnosis and prognosis of CRC.UALCAN and HPA databases were used to verify the difference of MCM6 expression in CRC and normal tissues,and explore its expression level changes in different clinical stages of CRC.The correlation between MCM6 and prognosis and clinicopathological features of CRC was analyzed by Cox regression analysis.Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis were performed to explore the potential functions of MCM6.To explore the MCM6 regulatory network and key genes,the STRING database was used to construct the protein-protein interaction(PPI)network.The correlation between MCM6 and infiltration of immune cells was analyzed based on the TIMER database.Results:MCM2-8 and MCM10 were significantly overexpressed in CRC tissues,having diagnostic significance for the assessment of CRC patients prognosis.High expression of MCM6 was associated with a better prognosis,having significant correlations with the clinical stage and clinicopathological features of CRC.The results of GO and KEGG pathway enrichment analysis suggested that MCM6 might be involved in biological processes such as cell cycle and RNA transport.MCM6 was also significantly corelated with infiltration level of various immune cells.Conclusion:The results showed that MCM6 is highly expressed in CRC,closely associated with the tumor development,prognosis,clinicopathological features,and tumor immune infiltration of CRC patients,and MCM6 might be a biomarker for the diagnosis and prognosis of CRC,as well as a potential therapeutic target for CRC.
