The influence of dysregulation of fatty acid metabolismon the progression of breast cancer

摘要 癌细胞持续增殖特性迫使其重调代谢以维持能量来源,目前认为代谢重编程是癌症标志之一。乳腺癌发病率连年增加,其代谢改变包括增强的糖酵解、三羧酸循环活性、谷氨酰胺分解和脂肪酸生物合成,在不同乳腺癌亚型中表现不同,并且与肿瘤生长、转移、死亡及耐药性等有着复杂关联。本文重点对目前乳腺癌脂肪酸代谢相关研究进行综述,以更好地理解脂肪酸代谢失调影响乳腺癌发展及其中相关分子机制,以期为针对代谢靶点的新型抗癌疗法提供线索。 Cancer cells'constant proliferation forces them to reset their metabolism to maintain energy sources,and metabolic reprogramming is now considered a hallmark of cancer.The incidence of breast cancer has increased year after year.Its metabolic changes include enhanced glycolysis,tricarboxylic acid cycling activities,glutamine decomposition and fatty acid biosynthesis,which have different manifestations in different breast cancer subtypes,and they also have complex associations with tumor growth,metastasis,death and drug resistance.This review focuses on the current research about fatty acid metabolism in breast cancer,so as to better understand how dysregulation of fatty acid metabolism regulates breast cancer progress,survive and produce the mechanism of tumor drug resistance,excepting to provide clues for new anti-cancer therapies targeting metabolic targets.
