摘要
目的探究化痰祛湿方对肥胖型多囊卵巢综合征(polycystic ovary syndrome,PCOS)大鼠的改善作用及对卵巢组织转化生长因子-β(transforming growth factor-β,TGF)-β/结缔组织生长因子(connective tissue growth factor,CTGF)通路的影响。方法选取45只SD雌性大鼠,其中随机取8只为对照组,其余大鼠给予来曲唑加高脂乳剂建立肥胖型PCOS模型。32只大鼠建模成功并随机分为4组,模型组,化痰祛湿方高、低剂量组及阳性组,每组各8只。低、高剂量组分别给予16.65 g/kg、83.25 g/kg化痰祛湿汤,阳性组给予二甲双胍0.3 g/kg。21 d后检测各组大鼠体质量,血清黄体生成素(luteinizing hormone,LH)、促卵泡生成素(folliclestimulating hormone,FSH)、睾酮(testosterone,T)、雌二醇(estradiol,E2)水平,卵巢组织病理学变化及TGF-β/CTGF通路m RNA及蛋白表达情况。结果与对照组比较,模型组大鼠血清LH、T水平上升,FSH、E2水平下降(P<0.05);卵巢组织TGF-β、CTGF m RNA与蛋白表达均上调(P<0.05);卵巢组织可见闭锁卵泡数增加,颗粒细胞层减少。与模型组比较,化痰祛湿方低、高剂量组及阳性组大鼠上述指标均有明显改善(P<0.05),且高剂量组与阳性组的改善效果优于低剂量组大鼠(P<0.05),两组大鼠卵巢内可见成熟卵泡。结论化痰祛湿方能够改善肥胖型多囊卵巢综合征,其作用机制可能与TGF-β/CTGF信号通路有关。
Objective To explore the improvement effect of Huatan Qushi recipe on obese polycystic ovary syndrome(PCOS)rats,and the effect of connective tissue growth factor(CTGF)pathway on the transforming growth factor(TGF)-β/CTGF pathway.Methods Forty-five SD female rats were selected,of which 8 were randomly selected as the control group,and the remaining rats were given letrozole plus high-fat emulsion to establish an obese PCOS model.Thirty-two rats were successfully modeled and randomly divided into 4 groups,model group,Huatan Qushi recipe high,low dose groups and positive group,with 8 rats in each group.The low-dose and high-dose groups were given 16.65 and 83.25g/kg Huatan Qushi recipe,respectively,and the positive group was given metformin 0.3g/kg after 21 days,the body weight,serum hormone levels[Luteinizing hormone(LH),follicle-stimulating hormone(FSH),testosterone(T),estradiol(E2)levels],ovarian histopathological changes and TGF-β/CTGF pathway mRNA and protein expressions were detected in each group.Results Compared with the control group,the levels of serum LH and T in the model group increased,while the levels of FSH and E2 decreased(P<0.05).The mRNA and protein expressions of TGF-βand CTGF in ovarian tissue were up-regulated(P<0.05).The number of atretic follicles increased and the granulosa cell layer decreased in ovarian tissue.Compared with the model group,the above indicators in the low-dose,high-dose and positive groups of Huatan Qushi recipe were significantly improved(P<0.05).And the improvement effect of the high-dose group and the positive group was better than that of the low-dose group(P<0.05),and mature follicles were found in the ovaries of the two groups.Conclusion Huatan Qushi recipe can improve obese polycystic ovary syndrome,its mechanism of action may be related to the TGF-β/CTGF signaling pathway.
