玫瑰茄活性成分的双水相系统提取及其抗氧化活性 被引量:1

Extraction of Active Components from Roselle Using Aqueous Two-phase System and Its Antioxidant Activity

摘要 为明确双水相系统对玫瑰茄活性成分的提取及其抗氧化活性的影响,本研究以玫瑰茄为材料,采用乙醇-(NH_(4))_(2)SO_(4)双水相系统提取玫瑰茄活性成分多酚和总黄酮。根据单因子测试结果,制定相应的测定方法,优化多酚和总黄酮的提取方法。进行体外抗氧化活性实验,研究了在最佳的提取工艺下玫瑰茄提取物DPPH自由基的清除率。结果显示,乙醇体积分数为40%,(NH_(4))_(2)SO_(4)质量浓度为0.24g/mL,液料比为40∶1(mL/g),提取温度41.5℃,提取时间26.5min,多酚和总黄酮提取量最高,依次为(22.93±0.02)mg/g和(39.27±0.01)mg/g,与预测值接近;玫瑰茄多酚及总黄酮提取物、维生素C清除DPPH自由基的IC_(50)值分别为21.76和30.02μg/m L,总抗氧化能力优于维生素C。本研究结果有助于实现玫瑰茄资源的综合高效利用。 In order to clarify the effect of aqueous two-phase system on the extraction of active components and antioxidant activity of roselle,polyphenols and total flavonoids were extracted by ethanol ammonium sulfate aqueous two-phase system from roselle.According to the results of single factor test,the determination method of response was formulated,and the extraction method of polyphenols and total flavonoids was optimized.The antioxidant activity experiment in vitro was carried out to study the DPPH radical scavenging rate of roselle extract under the best extraction process.The results showed that the volume fraction of ethanol was 40%,the mass concentration of(NH_(4))_(2)SO_(4) was 0.24 g/mL,the liquid-solid ratio was 40:1(mL/g),the extraction temperature was 41.5 C,the extraction time was 26.5 min,the extraction amount of polyphenols and total flavonoids was the highest,(22.93±0.02)mg/g and(39.27±0.01)mg/g respectively,which was close to the predicted value;the IC_(50) value of roselle extract and vitamin C for scavenging DPPH free radical was 21.76 and 30.02μg/mL,the total antioxidant capacity is better than vitamin C.The results of this study were helpful to realize the comprehensive and efficient utilization of roselleresources.
