摘要
目的研究辣木籽水提物对H1N1流感病毒诱导的小鼠病毒性肺炎的治疗作用。方法以H1N1流感病毒滴鼻诱导小鼠病毒性肺炎模型,C57BL/6小鼠随机分为正常组(NOR;正常饲养,不作任何处理)、模型组(MOD;正常饲养,灌胃纯净水)、奥司他韦阳性组(OSE;灌胃给药奥司他韦15 mg/kg)和辣木籽高、中、低剂量组(MOS-H、MOS-M、MOS-L;灌胃给药辣木籽水提物分别为20、10、5 mg/kg),采用不同剂量辣木籽水提物灌胃治疗后,检测小鼠体重、肺指数、肺组织病理形态、超微病变、肺部炎症因子TNF-α、IL-6水平。结果与正常组(NOR)相比,模型组(MOD)小鼠肺指数显著增大(P<0.05),肺部炎症病变加重,肺组织TNF-α和IL-6水平显著升高(P<0.01)。与模型组相比,辣木籽高、中、低剂量组(MOS-H、MOS-M、MOS-L)小鼠肺指数均有显著降低(P<0.05),肺部炎症病变显著减轻,肺组织TNF-α和IL-6水平也均有显著降低(P<0.01)。结论辣木籽水提物可减轻H1N1诱导的小鼠病毒性肺炎,为其在抗病毒性肺炎研究领域提供一定的实验证据。
Objective In this experiment,the therapeutic effects of aqueous extract of Moringa oleifera seeds on H1N1 influenza virus-induced viral pneumonia in mice was investigated.Methods The mouse viral pneumonia model was established by intranasal infusion of H1N1 influenza virus.C57BL/6 mice were randomly divided into normal group(NOR;normal feeding,no treatment),model group(MOD;normal feeding,gavage of pure water),oseltamivir positive group(OSE;gavage administration of oseltamivir 15 mg/kg),and moringa seeds at high,medium and low dose groups(MOS-H,MOS-M,MOS-L;gavage administration of moringa seeds aqueous extract 20 mg/kg,10 mg/kg,and 5 mg/kg,respectively).The body weight,lung index,lung histopathological morphology,ultramicroscopic lesions and lung inflammatory factors tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6)levels were measured after treating with different doses of Moringa oleifera seeds aqueous extract using gavage method.Results Compared with the normal group(NOR),the lung index of mice in the model group(MOD)increased significantly(P<0.05),the inflammatory lesions in the lung were aggravated,and the expression levels of TNF-αand IL-6 in the lung tissues increased significantly(P<0.01).Compared with the model group,the lung index of the mice in the high,medium and low dose groups(MOS-H,MOS-M,MOS-L)of Moringa oleifera seeds aqueous extract were significantly reduced(P<0.05),the inflammatory lesions in the lungs were reduced,and the expression levels of TNF-αand IL-6 in the lung tissues were also significantly reduced(P<0.01).Conclusion The study indicates that Moringa oleifera seed aqueous extract can significantly reduce H1N1-induced viral pneumonia in mice,which provide experimental evidence to antiviral pneumonia research field.
