摘要
以简单原料葡醛内酯,经过3步反应,对伞形酮类显色底物4-甲基-7-氧香豆素-β-D-葡萄糖苷酸进行了合成,目标产物中涉及的各步中间体,均通过核磁(^(1)H-NMR、^(13)C-NMR)等方法进行了分析表征,确定了合成工艺的可靠性。同时,通过病原微生物的荧光显色实验显示,4-甲基-7-氧香豆素-β-D-葡萄糖苷酸与大肠杆菌代谢酶结合反应后,显色效果较好,能够快速精准的检测出大肠杆菌。
The umbelliferone chromogenic substrate 4-methyl-7-oxocoumarin-β-D-glucuronide was synthesized from a simple raw material glucuronide through three-step reaction,the involved intermediates in each step were analyzed and characterized by nuclear magnetic resonance(^(1)H-NMR,^(13)C-NMR)and other means,and the reliability of the synthesis process was confirmed.At the same time,the fluorescence color development experiment of pathogenic microorganisms showed that the color development effect was good after the binding reaction of 4-methyl-7-oxocoumarin-β-D-glucuronide with the metabolic enzymes of Escherichia coli,and it could quickly and accurately detect E.coli.
