福建省泉州市水产品中副溶血性弧菌的质谱鉴定和聚类分析 被引量:1

Mass spectrometry identification and clustering analysis on Vibrio parahaemol yticus isolated from aquatic products in Quanzhou city,Fujian province

摘要 目的 了解福建省泉州市水产品中分离的副溶血性弧菌的毒力基因分布及同源性关系。方法 采用基质辅助激光解析电离飞行时间质谱(MALDI-TOF MS),对2020年分离自福建省泉州市水产品的59株疑似副溶血性弧菌菌株进行鉴定,利用MALDI-Biotyper数据库软件进行同源性聚类分析;采用荧光PCR法对毒力基因进行检测。结果 59株可疑菌株中只有46株鉴定为副溶血性弧菌,经同源性分析46株菌株之间蛋白组分特征差异比较大,可分为6大类别;经毒力基因鉴定,不耐热溶血素基因(tlh)检出率为100%,耐热性溶血素基因(tdh)和耐热性溶血素相关溶血基因(trh)均未检出。结论 MALDI-TOF MS质谱仪可用于副溶血性弧菌属水平的快速鉴定,福建省泉州市水产品存在一定程度的非致病性副溶血性弧菌污染,菌株同源性较低。 Objective To understand the virulence gene distribution and homology relation of Vibrio parahaemolyticus isolated from aquatic products in Quanzhou city of Fujian province.Methods Fifty-ninesuspected Vibrio parahaemolyticus strains isolated from aquatic products in Quanzhou city,Fujian province in 2020 were identified by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry(MALDI-TOF MS),and homology clustering analysis was carried out by using the MALDI-Biotyper database software;virulence genes were detected by fluorescence PCR.Results Only 46 of the 59 suspected strains were identified as Vibrio parahaemolyticus,and the differences in protein fraction characteristics between the 46 strains were relatively large after homology analysis and could be classified into 6 patterns.The similarity between each pattern was different.After identification of the virulence genes,the thermolabile hemolysin gene(tlh)was detected in 1oo%,and neither the thermostable direct hemolysin gene(tdh)nor the thermostable related hemolysin gene(trh)was detected.Conclusion MALDI-TOF MS can be used for rapid identification of Vibrio parahaemolyticus at the genus level,and a certain degree of non-pathogenic Vibrio parahaemolyticus contamination exists in aquatic products in Quanzhou city of Fujian province,with low strain homology.
