摘要
优化5-氨基乙酰丙酸盐酸盐(5-ALA)醇质体的制备工艺。选用大豆卵磷脂及胆固醇作为囊材,采用乙醇注入法制备5-ALA醇质体。以热敏性、包封率、变形性、粒度分布等为评价指标,分别采用单因素实验和正交实验优化工艺条件。最优制备条件为:m_((大豆卵磷脂))∶m_((胆固醇))=9∶1,m_((囊材))∶m_((药物))=23.3∶1,无水乙醇加入体积为10mL,PBS溶液的pH值为7.4,搅拌时间为30min,超声时间为4min。本实验优化得到的5-ALA光动力-热敏醇质体包封率较高,变形性较好,粒度分布均匀。
To optimize the preparation conditions of 5-aminolevulinic acid hydrochloride(5-ALA)ethosomes.Soybean lecithin and cholesterol were used as capsule materials to prepare 5-ALA ethosomes by ethanol injection method.Taking thermal sensitivity,encapsulation ratio,deformability and particle size distribution as evaluation indexes,the process conditions were optimized by single factor test and orthogonal test.The optimum preparation conditions were as follows:m_((soybean lecithin))∶m_((cholesterol))=9∶1,m_((capsule material))∶m_((main drug))=23.3∶1,the volume of anhydrous ethanol added was 10mL,the pH of PBS solution was 7.4,the stirring time was 30min,and the ultrasonic time was 4min.The optimized 5-ALA photodynamic-thermosensitive ethosomes obtained in this experiment had high encapsulation efficiency,good deformability and uniform particle size distribution.
