不同处理对菥蓂种子萌发特性的影响

摘要 菥蓂作为传统的中藏药材,兼具医疗价值和商业价值。为探究解除菥蓂种子休眠、促进种子萌发和幼苗生根的最佳策略,本研究对野生菥蓂种子进行低温、GA_(3)处理,基本培养基的筛选和浓度激素的配比试验。结果表明,菥蓂种子存在休眠,在低温下和外源施加GA_(3)可以有效解除其休眠,其萌发率与低温处理时间、GA_(3)浓度呈正相关,4℃低温处理20 d发芽率达99.93%;GA_(3)浓度为400 mg/L时,浸种30 min发芽率达100%,根长为(3.91±0.49)cm,相较于ck分别提高和增长了84.5%和2.94 cm。菥蓂在1/2MS培养基中发芽率最高,为(55.27±4.07)%,在此基础上添加不同激素,获得了适合种子萌发的激素配比方案:1/2MS+0.5 mg/L GA3+0.5 mg/L NAA+1 mg/L 6-BA,发芽率高达91.67%;促进根系伸长的激素配比方案:1/2MS+1 mg/L NAA+0.5 mg/L 6-BA。 Thlaspi arvense L.as a traditional Tibetan medicine with both medical and commercial value.In order to explore the best strategy to relieve the dormancy of Thlaspi arvense L.seeds and promote seed germination and seedling rooting,in this study the wild Thlaspi arvense L.seeds were treated with low temperature and gibberellin,screened the basic medium and tested the ratio of concentration hormones.The results showed that Thlaspi arvense L.seeds had dormancy and could be effectively released under low temperature and exogenous application of gibberellin.The germination rate was positively correlated with low temperature treatment time and gibberellin concentration.The germination rate reached 99.93% after 20 days of low temperature treatment at 4℃.When the gibberellin concentration was 400 mg/L,the germination rate reached 100% after soaking for 30 min,and the root length was(3.91±0.49)cm,which was 84.5% and 2.94 cm higher than that of ck,respectively.The germination rate of Thlaspi arvense L.was the highest in 1/2MS medium,which was(55.27±4.07)%.On this basis,different hormones were added to obtain the hormone ratio scheme suitable for seed germination was 1/2MS+0.5 mg/L GA 3+0.5 mg/L NAA+1 mg/L 6-BA,the germination rate up to 91.67%.The hormone ratio scheme for promoting root elongation was 1/2MS+1 mg/L NAA+0.5 mg/L 6-BA.
