目的探讨miR-525-5p和结肠癌转移关联基因1(MACC1)mRNA水平在结肠癌组织中的表达,分析二者与结肠癌患者临床病理学特征的关系,为结肠癌的诊断和治疗提供新思路.方法选取行结肠癌根治术的结肠癌患者50例为观察组,同期选取结肠良性病变患者50例为对照组.收集患者的结肠组织标本与临床资料,利用实时荧光定量PCR(RT-qPCR)检测两组标本中miR-525-5p和MACC1 mRNA表达水平.采用独立样本t检验比较两组间miR-525-5p与MACC1 mRNA表达;采用Pearson相关分析结肠组织中miR-525-5p与MACC1 mRNA表达的相关性;利用受试者工作特征曲线(ROC)和曲线下面积(AUC)分析miR-525-5p与MACC1 mRNA表达在结肠癌诊断中的应用价值.结果RT-PCR结果表明,观察组中miR-525-5p的表达量显著低于对照组(P<0.05),MACC1 mRNA表达量显著高于对照组(P<0.05).Pearson相关分析显示,miR-525-5p与MACC1 mRNA在结肠癌组织中表达呈负相关关系(r=-0.779,P<0.05),miR-525-5p在结肠癌组织中的表达与结肠癌患者肿瘤分化程度、TNM分期有关(P<0.05),MACC1 mRNA与肿瘤分化程度和淋巴结转移有关(P<0.05).ROC工作曲线结果表明,miR-525-5p诊断结肠癌的曲线下面积(AUC)为0.783(95%CI:0.693~0.873),特异度为70%,灵敏度为76%.MACC1 mRNA表达诊断结肠癌的AUC为0.706(95%CI:0.605~0.807),特异度为90%,灵敏度为66%.结论结肠癌组织中miR-525-5p低表达及MACC1 mRNA高表达可能成为结肠癌患者潜在的风险评估指标.

Objective To investigate the expression of miR-525-5p and colon cancer metastasis associated gene 1(MACC1)mRNA in colon cancer tissues,and analyze the relationship between the expression of miR-525-5p and the clinicopathological characteristics of patients with colon cancer,so as to provide new ideas for the diagnosis and treatment of colon cancer.Method 50 patients with colon cancer who underwent radical resection of colon cancer were selected as the observation group,and 50 patients with benign colon lesions were selected as the control group at the same time.The colonic mucosa and clinical data of patients were collected,and the mRNA levels of miR-525-5p and MACC1 mRNA of specimens in the two groups were detected by real-time fluorescent quantitative PCR(RT-qPCR),and the expression levels of MACC1 mRNA protein in the colonic tissues were detected by immunohistochemical method.Independent sample t-test was used to compare the mRNA expression difference between miR-525-5p and MACC1 mRNA,and the relationship between them and the clinicopathological characteristics of patients with colon cancer.Pearson correlation analysis was used to examine the correlation between miR-525-5p and MACC1 mRNA expression in colon tissue.The clinical value of miR-525-5p and MACC1 mRNA in the diagnosis of colon cancer was analyzed by using ROC and AUC.Results RT-PCR results showed that the expression of miR-525-5p in the observation group was significantly lower than that in the control group(P<0.05),and the expression of MACC1 mRNA was significantly higher than that in the control group(P<0.05).The immunohistochemical results showed that the positive expression of MACC1 mRNA protein in the observation group was significantly higher than that in the control group(P<0.05).Pearson correlation analysis showed that miR-525-5p was negatively correlated with MACC1 expression in colon cancer tissues(r=-0.779,P<0.05).The expression of miR-525-5p in tissues was related to tumor differentiation and TNM stage of colon cancer patients(P<0.05),and MACC1 mRNA was related to tumor differentiation and lymph node metastasis(P<0.05).ROC working curve results showed that the area under curve(AUC)of miR-525-5p for diagnosis of colon cancer was 0.783(95%CI:0.693~0.873),with specificity of 70%and sensitivity of 76%.The AUC of MACC1 mRNA for diagnosis of colon cancer was 0.706(95%CI:0.605~0.807),the specificity was 90%,and the sensitivity was 66%.Conclusion The low expression of miR-525-5p and high expression of MACC1 mRNA in colon cancer tissues may be used as potential risk assessment indicators for colon cancer patients.
