采用HPLC法测定温敏水凝胶中白藜芦醇的含量

摘要 为建立白藜芦醇温敏凝胶的检测分析方法,采用高效液相法测定温敏凝胶剂型中白藜芦醇的药物质量浓度,选用色谱柱为Waters Symmetry C_(18)(4.6 mm×150 mm,5.0μm),柱温30℃,流动相为甲醇、水(体积比58∶42),流速为0.8 mL·min^(-1),检测波长为306 nm,进样量为10μL,以保留时间定性、峰面积定量检测白藜芦醇质量浓度。结果表明:白藜芦醇在25~500μg·mL^(-1)范围内线性程度良好(R^(2)=0.9994),稳定性相对标准偏差(RSD)值为1.08%,重复性精密性RSD为1.81%,精密度考察RSD值为1.17%,加样回收率为100.43%(RSD为2.41%,n=9)。该方法具有简便、特异性强、稳定性好、重现度高等优点,可为白藜芦醇温敏凝胶的检测提供依据。 In order to establish a detection and analysis method for resveratrol thermosensitive gel,the drug mass concentration of resveratrol in thermosensitive gel formulation was determined by HPLC.The chromatographic column was Waters Symmetry C_(18)(4.6 mm×150 mm,5.0μm),the column temperature was 30℃,the mobile phase was methanol and water(volume ratio 58∶42),the flow rate was 0.8 mL·min^(-1),the detection wavelength was 306 nm,and the injection volume was 10μL.The mass concentration of resveratrol was determined qualitatively by retention time and quantitatively by peak area.The results showed that the linearity of resveratrol was good in the concentration range of 25~500μg·mL^(-1)(R^(2)=0.9994),the RSD of stability was 1.08%,the RSD of repeatability and precision was 1.81%,the RSD of precision was 1.17%,and the recovery was 100.43%(RSD was 2.41%,n=9).The method is simple,specific,stable and reproducible.It can provide a basis for the detection of resveratrol thermosensitive gel.
