基于高通量测序的椭圆食粉螨体内微生物群落结构及多样性分析

摘要 目的:阐明椭圆食粉螨体内微生物群落结构及多样性。方法:实验室饲养椭圆食粉螨,利用Illumina MiSeq技术对其体内细菌16S rRNA基因的V4区和真菌ITS1序列进行测序,统计体内微生物的操作分类单元(OTUs)数量,分析其体内微生物群落结构及Alpha多样性。结果:获得椭圆食粉螨体内细菌16s rRNA和真菌ITS1有效序列209643和145646条,根据序列相似性>97%进行聚类分析,分别得到了248和146个OTUs。基于OTUs进行物种注释,共注释到细菌类群20门,32纲,110目,177科,128属;共注释到真菌类群3门,15纲,36目,65科,92属。其中,在门分类水平,本实验室饲养椭圆食粉螨体内细菌群落主要注释到厚壁菌门Firmicutes(43.52%)、拟杆菌门Bacteroidota(31.94%)、放线菌门Actinobacteriota(20.04%)、变形菌门Proteobacteria(4.40%);真菌类群以子囊菌门Ascomycota(98.60%)为绝对优势菌门。在属分类水平,细菌类群主要注释到葡萄球菌属Staphylococcus(43.27%)、土壤杆形菌属Solitalea(31.81%)、棒杆菌属Corynebacterium(7.61%)、考克氏菌属Kocuria(7.06%)、短杆菌属Brevibacterium(4.11%)、鞘氨醇单胞菌属Sphingomonas(2.45%);真菌优势属为Trichomonascus(71.52%)、曲霉属Aspergillus(10.38%)、链格孢属Alternaria(9.29%)。Alpha多样性指数表明椭圆食粉螨体内微生物群落结构具有较高的丰富度和多样性。结论:椭圆食粉螨体内细菌和真菌群落具有较高的多样性和丰富度,且个体间微生物多样性存在差异。 Objective:To investigate the microbial community structure and diversity in the body of Aleuroglyphus ovatus(A.ovatus).Methods:A.ovatus was fed in laboratory.The V4 region of the bacterial 16S rRNA gene and the fungal internal transcribed spaceR1(ITS1)region were sequenced with Illumina MiSeq.The numbeRof operational taxonomic units(OTUs)of microorganisms in the body was counted,and the microbial community structure and alpha diversity was analyzed.Results:A total of 209643 valid sequences of bacterial 16s rRNA and 145646 of fungal ITS1 were obtained.ClusteRanalysis was performed with threshold of 97%sequence similarity,yielding 248 and 146 OTUs respectively.The total OTUs of bacteria were annotated into 20 phyla,32 classes,110 orders,177 families and 128 genera,while the fungal OTUs were annotated into 3 phyla,15 classes,36 orders,65 families and 92 genera.At the phylum level,the bacterial communities in the body of A.ovatus fed in out laboratory mainly belonged to Firmicutes(43.52%),Bacteroidota(31.94%),Actinobacteriota(20.04%)and Proteobacteria(4.40%).The dominant fungi belonged to Ascomycota(98.70%).At the genus level,the bacterial communities mainly belonged to Staphylococcus(43.27%),Solitalea(31.81%),Corynebacterium(7.61%),Kocuria(7.06%),Brevibacterium(4.11%)and Sphingomonas(2.45%).The dominant fungi belonged to Trichomonascus(71.52%),Aspergillus(10.38%)and Alternaria(9.29%).Alpha diversity indexes indicated that the microbial community structure in A.ovatus had high abundance and diversity.Conclusion:The bacterial and fungal communities in A.ovatus have high diversity and abundance,and there are differences in microbial diversity among individuals.
