杠板归不同部位代谢组差异研究

摘要 目的:比较杠板归不同部位代谢物积累的差异,为揭示其不同部位化学成分与功效的相关性和制定杠板归质量评价方法提供参考。方法:利用非靶向代谢组学方法检测杠板归茎、叶、果中代谢物,采用主成分分析和正交偏最小二乘法-判别分析等多元统计分析方法筛选出差异代谢物(DAMs),利用京都基因与基因组百科全书(KEGG)平台对DAMs进行富集分析。结果:从杠板归茎、叶、果中共鉴定出442个成分,其中酒石酸、L-苹果酸、N-α-乙酰基赖氨酸、L-谷氨酰胺、2-氯-L-苯丙氨酸、哌啶酸、2-甲基丙醛、甜菜碱、2-苯乙醇、L-丝氨酸等为茎、叶、果中共有的优势成分。杠板归茎、叶、果的代谢物质差异显著,茎和叶、茎和果、叶和果的DAMs数量分别为724、573、583个。果中黄酮类成分橙皮素、金丝桃苷、高良姜黄素、根皮苷和紫杉醇的含量显著高于茎和叶,而木犀草素-7-葡萄糖苷、异鼠李素、黄芩素和槲皮素-3-O-葡萄糖苷含量显著低于茎和叶。茎、叶、果的DAMs主要富集在氨酰tRNA生物合成通路、精氨酸生物合成通路、乙醛酸和二羧酸代谢通路上。结论:杠板归茎、叶、果之间有大量DAMs,这对其药效可能具有重要影响,研究结果可为杠板归药材质量评价提供参考。 Objective:To compare the metabolite accumulation in different parts of Polygonum perfoliatum L.,so as to provide reference for revealing the correlation between chemical components and efficacy in different parts and formulating quality evaluation methods.Methods:The non-targeted metabonomic method was employed to detect the metabolites in the stems,leaves,and fruits of P.perfoliatum.Multivariate analysis methods such as principal component analysis and orthogonal partial least squares discriminant analysis were used for the screening of differential metabolites.The Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis was carried out for the differential metabolites.Results:A total of 442 substances were identified from the stems,leaves,and fruits of P.perfoliatum.Tartaric acid,L-malic acid,N-alpha-acetyllysine,L-glutamine,2-chloro-L-phenylalanine,pipecolic acid,2-methylpropanal,betaine,2-phenylethanol,and L-serine were common dominant components in stems,leaves,and fruits.Metabolites varied in the stems,leaves,and fruits.The number of differential metabolites of stem vs.leaf,stem vs.fruit,and leaf vs.fruit was 724,573,and 583,respectively.The fruits had higher content of hesperidin,hyperosid,norizalpinin,phlorizin,and paclitaxel and lower content of luteolin-7-glucoside,isorhamnetin,baicalein,and quercetin-3-O-glucoside than that the stems and leaves.The differential metabolites of stems,leaves,and fruits were mainly enriched in the aminoacyl-tRNA biosynthesis,arginine biosynthesis,and glyoxylate and dicarboxylate metabolism pathways.Conclusion:There are abundant differential metabolites between the stems,leaves,and fruits of P.perfoliatum,which may affect the efficacy.The findings can provide a theoretical basis for the follow-up quality evaluation of the medicinal materials of P.perfoliatum.
