过表达ERF76小黑杨的木材组分分析及影响 被引量:1

摘要 通过对过表达ERF76小黑杨木材的各项化学组分进行了相关检测并结合转录组数据FC对不同转基因株系化学组分存在的差异现象进行了相关解释并总结了FC与纤维素、半纤维素组分占比变化的规律。根据与野生型小黑杨(WT)化学组分差异大小,筛选得到ERF76过表达转基因株系(P1)并且比较其两年生与六年生木材化学组分变化,确定了ERF76基因过表达对化学组分的影响伴随植物生长发育并不断累积。针对组分变化的特点实行进一步实验。发现在碱蒸煮工艺处理后转基因木料得率较高。在氢氧化钠半纤维素提取实验中发现P1所含半纤维素中大多为聚合度较低的半纤维素。结合化学组分差异等因素从化学反应接触面积、木材本身性质两个方面揭示了转基因木料在碱法蒸煮工艺实验中具备高得率的原因。 The chemical components of the overexpressed ERF76 small black poplar wood were detected and the differences of the chemical components of different transgenic strains were explained in combination with the transcriptome data FC,and the law of the change of the proportion of FC with cellulose and hemicellulose components was summarized.According to the chemical composition difference with wild type small black poplar(WT),a transgenic strain(P1)with ERF76 overexpression was screened,and the chemical composition changes of wood at two and six years old were compared.It was determined that the influence of ERF76 overexpression on chemical composition accompanied with plant growth and development and continuously accumulated.Further experiments were carried out according to the characteristics of component changes.It was found that the yield of transgenic wood was higher after alkali cooking.In the extraction experiment of sodium hydroxide hemicellulose,it was found that the hemicellulose contained in P1 was mostly hemicellulose with low degree of polymerization.The reasons for the high yield of transgenic wood in alkaline cooking process were revealed from the contact area of chemical reaction and the properties of wood.
