玉米BBR-BPC基因家族全基因组鉴定及表达分析

Genome-wide Identification and Expression Analysis of Maize BBR-BPC Gene Family

摘要 对玉米BBR-BPC基因家族进行全基因组鉴定,分析基因结构、染色体分布和启动子顺式作用元件结合位点、分布和数量情况以及编码蛋白的理化性质、保守基序和蛋白二级三级结构、进化关系,基于转录组的结果研究基因的组织表达模式和高温、干旱等非生物胁迫以及外源生长素等诱导表达模式。结果表明,玉米BBR-BPC家族共有4个成员,家族基因的结构存在较大的差异,且存在可变剪切,共编码9个蛋白。定位分析发现,BBR-BPC1.1和BBR-BPC4定位在细胞核,其余除定位在细胞核,叶绿体中也有分布。BBR-BPC启动子区存在多种逆境胁迫、植物激素、组织部位特异表达、光响应等顺式作用元件。BBR-BPC成员在玉米不同材料、不同组织部位中的表达水平差异较大且受热、干旱等胁迫调控。在B73和Mo17材料中,BBR-BPC受热胁迫诱导表达,与qRT-PCR验证结果一致。研究结果表明,ZmBBR-BPC基因家族不同成员会在特定部位激活或者抑制相关基因表达,响应非生物胁迫。 The whole genome of maize BBR-BPC gene family was identified.The gene structure,chromosome distribution,distribution and quantity of cis-element binding sites of promoter,as well as the physical and chemical properties,conserved motifs,protein secondary and tertiary structure and evolutionary relationship of coding protein were analyzed.At the same time,we also investigated the tissue expression pattern and the induction of abiotic stresses such as high temperature,drought and exogenous growth factors based on the transcriptome results.The re⁃sults showed that there were four members of maize BBR-BPC family.There were great differences in the structure of family genes,with variable splicing,encoding 9 proteins.BBR-BPC1.1 and BBR-BPC4 were located in the nu⁃cleus,and the rest were located in the chloroplast in addition to the nucleus.There were a variety of cis-acting ele⁃ments in BBR-BPC promoter region,such as stress,phytohormone,specific tissue expression,light response and so on.The expression levels of BBR-BPC members in different materials and tissues of maize vary greatly,and were regulated by heat,drought and other stress.In B73 and Mo17 materials,BBR-BPC was induced by heat stress,which was consistent with the results verified by qRT-PCR.The results showed that different members of ZmBBRBPC gene family can activate or inhibit the expression of related genes at specific sites in response to abiotic stress.
