和络泄浊方对缺血性肾脏病大鼠血管紧张素Ⅱ及肾间质纤维化的影响

Effects of Heluo Xiezhuo Prescription on the Expression of Angiotensin II and Renal Interstitial Fibrosis in Chronic Ischemic Nephropathy Rats

目的 观察和络泄浊方对缺血性肾脏病大鼠血管紧张素Ⅱ及间质纤维化的影响,探讨防治慢性缺血性肾脏病病变的机制。方法 SD雄性大鼠随机分为假手术组、模型组、药物组,每组10只。采用单侧肾动脉不全结扎建立模型。实验第14天处死大鼠,取肾组织做光镜,观察各组大鼠肾脏形态学改变;采用免疫组化法检测肾组织α平滑肌肌动蛋白(α-SMA)、聚合酶链反应(PCR)法检测肾组织血管紧张素Ⅱ(Ang Ⅱ)表达。结果 光镜下模型组和药物组大鼠的肾间质均有损伤。与假手术组比较,模型组和药物组肾小球硬化指数评分及肾小管间质损伤评分均明显升高(P<0.05),而药物组较模型组低(P<0.05)。药物组、模型组α-SMA、Ang Ⅱ表达均高于假手术组,且与模型组比较,药物组α-SMA、Ang Ⅱ表达下降(P<0.05)。结论 和络泄浊方可以通过下调α-SMA、Ang Ⅱ表达减轻缺血性肾脏的纤维化。

Objective To observe the effect of Heluo Xiezhuo prescription on chronic ischemic nephropathy rats expression of Ang II and renal interstitial fibrosis in chronic ischemic nephropathy rats,to explore the mechanism of prevention and treatment of chronic ischemic nephropathy.Methods Male SD rats were randomly divided into sham operation group,model group,drug group[Heluo Xie-zhuo prescription 14.0g/(kg·d)]by random number table method.The unilateral renal artery insufficiency ligation was used to estab-lish animal model.In the 14th day of experiment.The rats were put to death and nephridial tissue was collected to take light micro-scope examination,the changes of renal morphology in all group were observed.Immunohistochemistry was used to detect the expres-sion ofα-SMA,the mRNA expression of Ang II in nephridial tissue was detected by RT-qPCR.Results Under the light microscope,the glomerulus and tubulointerstitium in the model group and drug group had damage,compared with sham operation group,the in-flammatory disorders and fibrosis foci were displayed to different extents in the model group and drug group(P<0.05).Compared with model group,the expression ofα-SMA and Ang II in drug group was increased(P<0.05),which of both groups were higher than those of sham operation group(P<0.05).Conclusion Heluo Xiezhuo prescription can reduce Ang II andα-SMA expression,thereby pre-venting Interstitial fibrosis in ischemic nephropathy.
