摘要
【目的】革兰氏阴性菌细胞壁的主要成分是脂多糖(LPS),由于革兰氏阴性菌的感染会引发多种繁殖障碍疾病,降低家畜繁殖力。本试验旨在研究革兰氏阴性菌感染对母兔子宫颈IL-1β蛋白表达水平的影响。【方法】选取27只健康新西兰白兔,分为3个组,分别为0h组:3只兔不做处理,LPS组:12只兔耳缘静脉注射LPS(0.5mg·kg^(-1)),对照组:12只兔注射生理盐水(0.5mg·kg^(-1)),在注射后1.5、3、6、12h(n=3)收集子宫颈,采用免疫组化的方法测定子宫颈中IL-1β蛋白表达水平。【结果】母兔子宫颈中黏膜层和肌层均有IL-1β蛋白的表达。在黏膜层中,IL-1β蛋白表达量在注射1.5h后,LPS组极显著高于对照组(P<0.01),且对照组中1.5h显著低于其他时间点(P<0.05),LPS组各时间段无显著差异(P>0.05);在肌层中,IL-1β蛋白表达量在LPS处理后1.5h和6h极显著高于对照组(P<0.01),使用LPS处理该组12h后,其表达量出现显著升高(P<0.05),且对照组中各时间段无显著差异(P>0.05),LPS组6、12h显著高于3h(P<0.05)。【结论】LPS诱导了母兔子宫颈中IL-1β蛋白表达水平的显著增加,且在黏膜层和肌层有一定的差异性。
[Objective]Lipopolysaccharide(LPS)is the major pathogenic component of gram-negative bacteria.Female reproductive tract infection triggered lots of reproductive disorder diseases,which decreases reproduction ability.The aim of this study was to reveal the effects of LPS on the expression level of IL-1β protein in cervix of female rabbits.[Method]27 health New Zealand female rabbits,3 animals were not treated as 0h,12 rabbits were injected with LPS according to the dosage of 0.5mg/Kg as LPS group,and other 12 ones were injected with saline as control group.The cervixes were collected at 1.5,3,6 and 12h after injection.The expression level of IL-1β protein was examined by immunohistochemistrical assay(IHC assay).[Results]The results showed:The protein of IL-1β was expressed in the cervical mucosalayer and muscularis.In the mucosal layer,IL-1β protein level in LPS group was significantly increased at 1.5h compared with the control group(P<0.01).And IL-1β protein level at 1.5h in the control group was significantly lower than other time points within group(P<0.05),LPS group had no significant difference with time-course(P>0.05).In the muscle layer,the expression of IL-1β protein after LPS treatment at 1.5h and 6h was significantly higher than the control group(P<0.01),and its expression was significantly higher than the control group at 12h(P<0.05),and no significant difference was observed within the control group(P>0.05).The expression of IL-1β protein at 12h was significantly higher than 3h in the LPS group(P>0.05).[Conclusion]LPS induced a significant increase in the expression of IL-1βprotein in the cervix,and there were some differences in the mucosal layer and muscle layer.
