摘要
为了研究小麦TaCPP20-5B基因在小麦生长发育过程中的作用,对小麦中CPP(Cysteine-richpolycomb-like protein)家族基因进行了全基因组鉴定和生物信息学分析,并对该家族中的TaCPP20-5B基因进行了克隆、蛋白质理化性质和二级结构、基因表达量检测和亚细胞定位分析。结果表明,在小麦基因组中共鉴定出CPP家族成员26个,系统发育进化树共有A、B、C和D 4个亚族。小麦CPP家族基因在其他组织中表达量明显高于叶片,旱热胁迫后TaCPP13和TaCPP20表达量显著提升;TaCPP20-5B基因编码区全长为1083 bp,共编码360个氨基酸,蛋白质分子量为40.02 kD,不稳定系数为52.71,脂肪系数为57.69,总平均亲水性指数为-0.716,等电点为8。蛋白质二级结构分析表明,α-螺旋、β-折叠、无规则卷曲和直链延伸分别占比20.83%、3.61%、61.94%和13.61%。TaCPP20-5B在小麦叶片中表达量最低,在根和茎中表达量相近,是叶片的2.6~2.7倍。亚细胞定位发现TaCPP20-5B定位于细胞核中。本研究结果可为进一步研究TaCPP20-5B基因在小麦中的功能提供参考依据。
To investigate the role of T aCPP20-5B gene in wheat growth and development,genome wide identification and bioinformatics analysis were conducted on the CPP family genes in wheat,and cloning,protein physicochemical properties,secondary structure and gene expression of analysis,and subcellular localization analysis were performed on the TaCPP20-5B gene in this family.The results showed that a total of 26 members of the CPP family have been identified in the wheat genome,and the phylogenetic tree indicates that the family containing four subfamilies:A,B,C,and D.The expression levels of wheat CPP family genes were significantly higher in other tissues than in leaves,and the expression levels of TaCPP13 and TaCPP20 were significantly increased after drought and heat stress.The coding region of TaCPP20-5B is 1083 bp in length,encoding 360amino acids.The molecular weight,instability index,fat index and total average hydrophilicity index,isoelectric point of the protein is 40.02 kD,52.71,57.69,-0.716 and 8.Secondary structure analysis of proteins indicates that,α-helix,β-folding,random coil and linear extension accounted for 20.83%,3.61%,61.94%and 13.61%.The expression level of TaCPP20-5B was the lowest in wheat leaves,and the expression level of TaCPP20-5B was similar in roots and stems,which was 2.6 to 2.7 times that in leaves.Subcellular localization reveals TaCPP20-5B localization in the nucleus.The results of this study can provide a reference basis for further research on the function of the TaCPP20-5B gene in wheat.
