骨髓间充质干细胞对骨质疏松性椎体骨折大鼠疼痛行为改善的影响

摘要 目的 :探讨骨髓间充质干细胞(BMSCs)对骨质疏松性椎体骨折大鼠疼痛行为改善的影响。方法 :选取健康大鼠建立骨质疏松性椎体骨折模型,随机分为健康组、模型组、BMSCs低剂量组、BMSCs中剂量组、BMSCs高剂量组、尼尔雌醇组。健康组与模型组大鼠给予5 mL/kg生理盐水灌胃;BMSCs低剂量组、中剂量组和高剂量组分别通过尾静脉注射BMSCs 104个/mL、106个/mL和108个/mL,每周2次;尼尔雌醇组采用尼尔雌醇灌胃,0.15 mL/kg,每周1次,连续4周。自发痛测定机械痛阀(MWT)、热刺激爪退缩阈(TWL)检测自发痛行为(抬腿次数、抬腿持续时间);万能材料试验机检测力学性能,ELISA检测血清骨特异性碱磷酶(BALP)、Ⅰ型胶原C端异物肽(CTX-Ⅰ),H-E染色观察病理形态。结果 :与健康组比较,模型组、BMSCs低剂量组大鼠MWT、TWL和BALP以及最大载荷、最大应变、弹性模量降低,CTX-Ⅰ及抬腿次数、抬腿持续时间升高。与模型组、BMSCs低剂量组比较,BMSCs中剂量组、尼尔雌醇组MWT、TWL和BALP以及最大载荷、最大应变、弹性模量升高,CTX-Ⅰ及抬腿次数、抬腿持续时间降低。与BMSCs中剂量组、尼尔雌醇组比较,BMSCs高剂量组大鼠MWT、TWL和BALP以及最大载荷、最大应变、弹性模量升高,CTX-Ⅰ及抬腿次数、抬腿持续时间降低。结论:BMSCs对改善骨质疏松性椎体骨折大鼠的疼痛行为可能具有一定积极效用,在骨质疏松性椎体骨折的治疗中可发挥一定的保护作用。 Objective:To investigate the effect of bone marrow mesenchymal stem cells(BMSCs)on the improvement of pain behavior in rats with osteoporotic vertebral fracture.Methods:Healthy rats were selected,and were established to model osteoporotic vertebral fracture.They were randomly divided into a healthy group,a model group,a BMSCs low-dose group,a BMSCs medium-dose group,a BMSCs high-dose group,and a neilestrol group.The healthy group and model group were given 5 mL/kg normal saline intragastric administration.104 cells/mL,106 cells/mL and 108 cells/mL of BMSCs were injected into the rats in low-dose BMSCs group,mediumdose BMSCs group and high-dose BMSCs group through the tail vein separately twice a week.Rats in the nilestriol group was injected via the stomach with 0.15 mL/kg nilestriol once a week for 4 weeks.MWT and TWL were used to detect spontaneous pain behavior(number of leg lifting and duration of leg lifting).The mechanical properties were detected by universal material testing machine.Serum bone alkalin phophatase(BALP)and collagen C telopeptide typeⅠ(CTX-Ⅰ)were detected by ELISA,and the pathological morphology was observed by H-E staining.Results:Compared with the healthy group,MWT,TWL,BALP,maximum load,maximum strain,and elastic modulus of rats in model group and BMSCs low-dose group decreased,while CTX-Ⅰ,the number of leg lifting,and the duration of leg lifting increased.Compared with the model group and BMSCs low-dose group,MWT,TWL,BALP,maximum load,maximum strain,and elastic modulus were increased in BMSCs medium dose group and nierstradiol group,while CTX-Ⅰ,the number of leg lifting and the duration of leg lifting were decreased.Compared with the BMSCs medium dose group and nierstradiol group,MWT,TWL,BALP level,maximum load,maximum strain and elastic modulus were increased in BMSCs high-dose group,while CTX-Ⅰlevel,the number and duration of leg lifting were decreased.Conclusion:BMSCs may have a positive effect on improving the pain behavior of rats with osteoporotic vertebral fracture,suggesting that BMSCs could play a protective role in the treatment of osteoporotic vertebral fracture.
